Recent studies have demonstrated that androgenic stimulation of early and late responding genes depends on the nuclear uptake of the receptor-steroid complex. The longer the complex is retained in the nucleus, the shorter the lag period and the greater the magnitude of the response. Most antiandrogens or their metabolites bind to the cytosolic androgen receptor, but the relative receptor binding affinities do not strictly relate to their biological activities, since the former assay does not differentiate antagonists from weak agonists. Alternatively, androgen antagonists are believed to abrogate the action of androgenic steroids by interfering with the receptor function. However, the exact mechanisms by which antiandrogens bring about their actions remain to be established.

“Pure” antiandrogens of the cyproterone or flutamide type have no or only a transient effect on testes function and spermatogenesis. They are not useful for male contraception. Only antiandrogens with antigonadotrophic (progestational) activity or combinations of “pure” antiandrogens with a central inhibitory steroid inhibit spermatogenesis and fertility. The inhibitory effect of cyproterone acetate on spermatogenesis is a direct one as has been shown in animal experiments (rats). Most susceptible to cyproterone acetate are the accessory sexual glands, followed by the epididymis, the Sertoli cell and testes function. Concerning the use of cyproterone acetate as a male contraceptive it seems to be very difficult to find a dose sufficient to inhibit fertility without influencing libido.

Until 1972 it was shown that the androgen action in the prostatic tissue was through receptor proteins. In 1975 the synthesis of methyltrienolone (MT) opened a new field in searching receptor proteins. This synthetic androgen discriminates between receptors and other proteins such as TeBG and albumin. The main purpose in this investigation was to obiain the electrophoretic patterns of human prostatic androgen-binding proteins and compare them with those of other tissues from male genital tract (e.g., testes and epididymis), using as a reference the already described pattern in blood serum. The experimentation was carried out with cytosol (105,000 g) and nuclear fractions from the homogenized tissue. Both fractions were incubated with the tritiated steroids and then treated with dextran-coated charcoal. Polyacrylamide gel electrophoresis (PAGE) was then performed at pH 8.3 with acrylamide (T) 7.7%. 190, 0.455, 0.570, 0.660, and 1.055 and bound 3H-MT in Rf 1.01. We concluded from these results that the electrophoretic patterns in the prostate gland shared some radioactive areas resembling TeBG and albumin, although they present low binding capacity. The other peaks with different electrophoretic mobilities are in the process of further identification.
