Further characterization of marek's disease virus‐infected lymphocytes. I. In vivo infection

Abstract: Previous reports from this laboratory identified bursaderived lymphocytes (B cells) and non-B cells as the predominant cell types respectively involved in the early cytolytic and subsequent latent infection of chickens with Marek's disease virus (MDV). It was not known whether these differences were qualitative or quantitative or if the method for detection of latent infection (viral antigen production after 48 h of in v i h o cultivation) was sensitive enough. To further define the cells involved in the vario… Show more

“…Cloned MDV isolates CU-2 (low oncogenicity) and JM-16 (moderate oncogenicity) have been described (Smith & Calnek, 1973;Calnek et al, 1984). All inocula for these experiments were from stocks of infected chicken kidney cell cultures stored at -196 °C.…”

“…Spleens were collected aseptically and processed as reported by Calnek et al (1984). Briefly, spleen cells were gently forced through an autoclavable 60 ~tm mesh (Tetco, Elmsford, N.Y., U.S.A.), washed once in phosphate-buffered saline (PBS) pH 7.3 and centrifuged over Ficoll-Paque (Pharmacia) at 400 g for 20 min.…”

“…By comparison, resistance in line 6 1 (B2 MHC haplotype) is strongly associated with the MDV-1 locus (see earlier). It has been postulated that resistance resides at the level of target cell numbers and their susceptibility to MDV infection and is evident during the lytic phase of infection (16,32).…”
