Functional Role and Therapeutic Potential of the Pim-1 Kinase in Colon Carcinoma

Abstract: We demonstrate that Pim-1 plays a pivotal role in several tumor-relevant signaling pathways and establish the functional relevance of Pim-1 in colon carcinoma. Our results also substantiate the RNAi-mediated Pim-1 knockdown based on polymeric polyethylenimine/small interfering RNA nanoparticles as a promising therapeutic approach.

“…This includes the downregulation of Cyclins B1 and D1 (deceleration of cell cycle in the transition from G2 to M and from G1 to S, respectively), the activation of caspase-3/-7 (induction of apoptosis) and the decrease in the pro-survival protein Survivin. The latter finding supports our previous studies in colon carcinoma [39, 43] suggesting a SATB1 – Pim1 – Survivin axis that leads to a parallel Survivin decrease upon SATB1 knockdown. However, we found Pim1 protein levels being even elevated instead.…”

“…3,18,24,30,32 Other genes affected include Pim-1, which we recently demonstrated to be functionally relevant in colon carcinoma and to play a pivotal role in several signaling pathways. 33 Here, SATB1 knockdown led to a 50% reduction in Pim-1, and we have shown previously that this leads to major changes in oncogenic signal transduction with regard to p21 Cip1/WAF1 , STAT3, JNK, c-Myc and survivin, and in the levels of apoptosis-related proteins Puma, Bax and Bcl-xL. 33 Thus, oncogenic SATB1 effects may also be mediated through Pim-1 and go all the way down to survivin, which is highly expressed in various tumor entities including colon carcinoma and is associated with elevated cell survival and proliferation, higher tumor grade, poor prognosis as well as resistance to chemotherapy and radiation (for review, see Ref.…”

“…PIM1 phosphorylation has also been linked to the development of chemoresistance in hypoxic conditions via regulation of the mitochondrial transmembrane potential in pancreatic, cervical and colon cancer cells (38, 39). Inhibition of PIM kinases in prostate, breast and colon cancer decreases tumor cell proliferation while increasing apoptosis (40, 41). Interestingly, PIM kinases have not previously been implicated in MPNST pathogenesis.…”
