Functional Implications of the IL-6 Signaling Pathway in Keloid Pathogenesis

Abstract: The molecular mechanism(s) behind keloid pathogenesis remains unclear. Previously by global gene expression analysis of keloid fibroblasts (KFs), we implicated the IL-6 signaling pathway in keloid pathogenesis. Here, we determine a functional role of IL-6 signaling in keloid scars. Primary cultures of KFs and surrounding nonlesional fibroblasts (NFs) were subjected to induction or inhibition of IL-6 or its specific receptor IL-6 receptor alpha (IL-6R alpha) and detection of their effects on extracellular matri… Show more

“…[5][6][7][8][9][10][11][12] Modifications in growth factor-regulated cell growth and apoptosis, intracellular signal transduction, and epidermis-dermis interactions have been reported. [13][14][15][16][17][18][19][20][21] Changes in the level of genes involved in the above phenomena were also detected in gene profiling studies. 22,23 Despite these findings, knowledge-based specific approaches for prevention and efficacious treatment of keloids are still absent, and the lack of proper in vitro and animal models that recapitulate the pathophysiology of keloids further hampers progress.…”

“…Il6 activity has also been implicated in keloid formation. 32 In the inflammation category, we identified 25 genes with a 4-fold increase in the thermal stress areas and eight genes that showed a 4-fold, or greater, decrease in expression. These results are shown in Table 4.…”

“…Ostatnio postuluje się, że IL-6 odgrywa kluczową rolę w tego typu procesach, dlatego hamowanie tej cytokiny może stanowić obiecujący cel terapeutyczny w zapobieganiu powstawaniu blizn [23,24]. Terapia światłem LED obniża poziom mRNA IL-6, co w konsekwencji może przeciwdziałać bliznowaceniu miejsc poddanych zabiegom, po urazach czy w trądziku [4,25].…”
