Functional genomics of hsp-90 in parasitic and free-living nematodes

Abstract: Heat shock protein 90 (Hsp-90) is a highly conserved essential protein in eukaryotes. Here we describe the molecular characterisation of hsp-90 from three nematodes, the free-living Caenorhabditis elegans (Ce) and the parasitic worms Brugia pahangi (Bp) and Haemonchus contortus (Hc). These molecules were functionally characterised by rescue of a Ce-daf-21 (hsp-90) null mutant. Our results show a gradient of rescue: the C. elegans endogenous gene provided full rescue of the daf-21 mutant, while Hc-hsp-90 provid… Show more

“…In order to confirm the involvement of DAF-21 in the observed phenotypes, we reduced DAF-21 levels in wt worms by feeding RNA interference (RNAi) using a construct described before [14], [28]. Worms remained sterile as previously observed [13][15]. We used worms at day 4 and day 7 after hatching and measured the frequency of thrashing (Table 1).…”

“…Daf-21 is an essential gene, as observed by RNAi and knockout strains [15]. Given the essential nature of DAF-21, we were intrigued by the mild phenotype of the daf-21(p673) nematodes.…”

“…These pathways have also been implicated in development, metabolism and behaviour in other phylogenetically distant free-living nematodes, APNs (Ogawa et al , 2009; Bento et al , 2010; Albarqi et al , 2016) and in the PPN B. xylophilus (Zhao et al , 2013). Other genes from the DAF pathway have been reported to be necessary for parasitism, such as DAF-21 / HSP90 (Birnby et al , 2000; Gillan et al , 2009; Lourenço-Tessutti et al , 2015). This finding raises the question of whether other DAF genes are determinants at different steps during the parasitic nematode life cycle.…”

“…The greater efficiency of complementation by Hc-daf-16.2 may reflect the closer phylogenetic relationship between C. elegans and H. contortus (both within clade V), compared with the relatively distant relationship of C. elegans and S. stercoralis (a member of clade IV (Blaxter et al, 1998). A similar correlation between cross-species complementation efficiency and phylogenetic relatedness is seen in the case of parasite orthologues of the heat shock protein 90 (HSP-90) encoded by C. elegans daf-21 (Gillan et al, 2009). These authors found that the H. contortus orthologue Hc-hsp-90 can partially complement a daf-21 mutation in C. elegans , whereas the orthologue Bm-hsp-90 from the filarial nematode, B. malayi (a member of nematode clade III (Blaxter et al, 1998) cannot.…”
