Functional Genomics Identified Novel Genes Involved in Growth at Low Temperatures in Listeria monocytogenes

Abstract: Listeria monocytogenes is a Gram-positive pathogen that contributes to foodborne outbreaks due to its ability to survive at low temperatures. However, the genetic determinants of Lm involved in growth at low temperatures have not been fully defined.

“…In addition, ribosome-related genes are also frequently identified under extreme environmental conditions (including acidity, low temperature, and high pressure, etc.) [ 19 , 36 , 37 ]. In detail, in the CC category, the most significantly enriched GO terms were “ribosome”, “intracellular ribonucleoprotein complex”, and “ribonucleoprotein complex” in the up-regulated genes ( Figure S1A ) and “bacterial-type flagellum” and “cell projection” in the down-regulated genes ( Figure S1D ).…”

“…Sample libraries were prepared and analyzed by barcoded Tn-seq as described previously. 41 Library preparation and sequencing were performed on the Illumina HiSeq PE150 platform (Personalbio, Shanghai, China), generating an average of 1 G high-quality sequencing reads per sample. This experiment was performed in triplicate.…”

“…The high-density transposon mutant library (containing 63,666 unique insertion mutants) was constructed from previous studies in our laboratory ( 36 , 37 ). Briefly, the temperature-sensitive plasmid pGPA2 was first electroporated into competent cells of L. monocytogenes .…”
