Functional Consequences of the Developmental Arrest and Follicular Exclusion of Anti-Double-Stranded DNA B Cells

Abstract: Anti-dsDNA B cells are actively tolerized in nonautoimmune BALB/c mice, as manifested by their developmental arrest, follicular exclusion, and rapid turnover rate. Previously, we have documented changes in the maturation status and follicular localization of anti-dsDNA B cells in autoimmune-prone MRL (+/+ and lpr/lpr) mice. To determine whether these differences in developmental status and follicular localization affect the functional capacity of anti-dsDNA B cells, we have now compared their in vivo life span… Show more

“…By contrast, BCR-mediated upregulation of CD86 expression is fully functional in insulin-specific B cells from V h 125×V κ 125 mice 27 , yet these cells are unable to mount T-cell-dependent immune responses 26 . Interestingly, all of these anergic B cells, including those from HEL-, Ars-, ssDNA-, insulin-and dsDNA-specific models, can be stimulated by IL-4 and CD40-specific antibody in vitro, suggesting that they are capable of responding to T-cell-derived signals if they are provided in the correct context 27,33,41,65 (S.B.G. and J.C.C., unpublished observations).…”

“…The pairing of the VH3H9 IgH with the V 1 L chain generates an anti-dsDNA Ab (27,28). This facilitates tracking of anti-dsDNA B cells in vivo in a diverse repertoire by the use of anti-Ig reagents (17)(18)(19).…”

“…B cells that fail to edit successfully away from their autoreactive specificity undergo apoptosis in the bone marrow, prohibiting further differentiation (4,8). Other autoreactive B cells exit the bone marrow and either developmentally arrest at a transitional stage in the spleen (9,10) or become anergic B-2 cells (5,11,12).…”
