Functional characterization of cell-wall-associated protein WapA in Streptococcus mutans

Abstract: Streptococcus mutans is known as a primary pathogen responsible for dental caries. One of the virulence factors of S. mutans in cariogenicity is its ability to attach to the tooth surface and form a biofilm. Several surface proteins have been shown to be involved in this process. A 29 kDa surface protein named wall-associated protein A (WapA, antigen A or antigen III), was previously used as a vaccine in animal studies for immunization against dental caries. However, the function of WapA in S. mutans is still … Show more

“…in vivo and in situ conditions and has been used to determine the mechanical properties and molecular forces of bacterial cells Schär-Zammaretti & Ubbink, 2003a, b;van der Mei et al, 2000;Zhu et al, 2006). “…Mechanical properties of macromolecules at the microbial surface are commonly probed by studying the tip-cell interactions observed in force spectroscopy experiments (van der Aa et al, 2001). Tip-cell-surface transient and local interactions serve as a reasonable model for probing adhesive properties at the surface of microbial cells (Pelling et al, 2005;van der Aa et al, 2001;Zhu et al, 2006). This Table 1.…”

“…Interestingly, the expression of one ortholog of spxA (SMU.2084c) that encodes a transcriptional regulator known to positively regulate genes in response to oxidative stress (41) was increased about 1.5-fold, whereas SMU.1548c, encoding a histidine kinase of a TCS, was downregulated approximately 30%. Interestingly, the ackA mutant exhibited a 2-fold increase in the level of transcription of wapA (SMU.987), encoding a cell wall-associated protein implicated in sucrose-independent aggregation and biofilm formation (42). Previously, a wapA mutant of S. mutans UA140 was shown to have substantially impaired biofilm formation under low-sucrose conditions (42).…”

Previously, a wapA mutant of S. mutans UA140 was shown to have substantially impaired biofilm formation under low-sucrose conditions (42). These changes in expression of wapA and spx are thus consistent with the observed alterations in biofilm formation and oxidative stress tolerance, respectively, by the ackA mutant.…”

“…To validate the results of the microarray data, 10 randomly selected genes (Table 2) were subjected in triplicate to quantitative RT-PCR utilizing Sybr green detection in a Rotor Gene 6000 (Corbett) (75). For each sample (WT, ⌬310 mutant, and WT/Zn 2ϩ ), 2 g of total RNA was used for first-strand cDNA synthesis with a commercial RT kit (Promega) as recommended by the manufacturer, and the primers (Table 2) were designed according to the published genomic sequence of 05ZYH33 (8).…”
