Functional characterization of a glutamate/aspartate transporter from the mosquitoAedes aegypti

Abstract: SUMMARYGlutamate elicits a variety of effects in insects, including inhibitory and excitatory signals at both neuromuscular junctions and brain. Insect glutamatergic neurotransmission has been studied in great depth especially from the standpoint of the receptor-mediated effects, but the molecular mechanisms involved in the termination of the numerous glutamatergic signals have only recently begun to receive attention. In vertebrates, glutamatergic signals are terminated by Na+/K+-dependent high-affinity excit… Show more

“…Electrophysiological properties of AaePAT1 were determined as previously described (Umesh et al, 2003) using a two electrode voltage clamp (TEV-200, Dagan Instruments, Minneapolis, MN, USA) interfaced to a computer with a Digidata 1200 A/D controller using the pCLAMP 6.0 program suite (Axon Instruments, Union City, CA, USA). Amino acid substrates were applied over a 9 s exposure in 150 μl volumes using a Rheodyne injection valve.…”

“…A total of more than a dozen Na + -coupled amino acid transporters have been cloned by Gill's group (Umesh et al, 2003), Boudko and Harvey's group and others (reviewed by Boudko et al, 2005b). When expressed in Xenopus oocytes, the five NATs from mosquito larvae exhibited characteristic profiles for uptake of the 20 structural amino acids.…”
