Functional and Epigenetic Studies Reveal Multistep Differentiation and Plasticity of In Vitro-Generated and In Vivo-Derived Follicular T Helper Cells

Abstract: Summary Follicular T helper cells (Tfh) provide critical help to B cells for germinal center (GC) formation. Mutations affecting SLAM-associated Protein (SAP) prevent GC formation due to defective T-B cell interactions, yet effects on Tfh cell differentiation remain unclear. We describe the in vitro differentiation of functionally-competent “Tfh-like” cells that expressed Interleukin-21, Tfh markers, and Bcl6, and rescued GC formation in SAP-deficient hosts better than other T helper (Th) cells. SAP-deficient … Show more

“…5B). These data are consistent with reduced expansion of GC B cells in these mice because multiple observations suggest that T FH cell development is a limiting factor for the generation and expansion of GC B cells (35)(36)(37). In contrast, MyD88 deletion in B cells resulted in little or no reduction in total T FH cell numbers following CpG-NPCGG immunization (Fig.…”

“…For example, adoptive transfer of WT in vitro-differentiated T FH -like cells into WT mice increased the magnitude of the GC response, indicating that the numbers of T FH cells can be limiting for GC reactions (35)(36)(37).…”

“…5a). However, their induction or co-expression in opposing subsets, especially with FOXP3, indicates that counter-regulation of their expression is incomplete 37,117 . Thus, these so-called master transcription factors behave less like lineage-definers and more like executors, expressed in response to environmental cues to carry out the induction of a defined set of gene effectors.…”

“…44). CD4 + T-cell lineage commitment is determined by the cytokine composition of the microenvironment in which the CD4 + T cells are primed and boosted via transcriptional regulation of lineage-specific cytokine gene families (38,39,45). This observation leads to the sobering possibility that CD4 + CCR5 + T cells (i.e., Th17 cells) responding to an HIV vaccine designed to elicit a persistent and protective antibody response could blunt protection or, worse, increase susceptibility to infection, as suggested in the adenovirus serotype 5 (Ad5) vaccine trials (13)(14)(15)(16).…”
