Functional Analysis of the Proximal 5′-Flanking Region of the N-Methyl-D-aspartate Receptor Subunit Gene, NMDAR1

Abstract: The NMDAR1 receptor subunit is a common subunit of N-methyl-D-aspartate receptors. We have previously characterized 3 kilobases (kb) of 5'-flanking sequence of the NMDAR1 gene and now report on the ability of this region to direct transcription of a reporter gene and on its interaction with nuclear proteins. The sequence 356 base pairs (bp) 5' of the first nucleotide of codon 1 was sufficient to express a luciferase reporter gene in rat PC12 pheochromocytoma cells. Additional sequences upstream of nucleotide -… Show more

“…However non-hormone response element containing genes, such as the AP1 response element (Paech et al 1997), can also respond to the estrogen receptor ligand (Katzenellenbogen et al 1996). An SP1 motif, which is similar to the AP1 motif, has been found in the NMDA receptor subunit 1 (Bai and Kusiak 1993;Bai and Kusiak 1995) and 2B (Suchanek et al 1995) promoters. Nevertheless, further investigations with gel shift and DNA footprinting assays are required to clearly answer this potential mechanism of action.…”

“…Co-transfection of luciferase and ␤-galactosidase constructs and assay of reporters were done as described previously (37,38). In some experiments, cells were used for RNA extraction 2 days after co-transfection.…”

“…DNA-Protein Interaction Analysis-Crude nuclear extractions were prepared from cells as previously described (37). DNA sequences used as probes in electrophoretic mobility shift assays (EMSA) are as follows: GC-box-a, 5Ј-GCCTCCGGGGTGGGCGACTGC; GC-box-b, 5Ј-CCGGG-GAGCGGGCGGAGAGCGTGGT; GC-Box-c, 5Ј-GCATCCTGGGCGGG-TGTGTGC.…”

“…PC12 pheochromocytoma cells were cultured in DMEM supplemented with 5% FCS and 5% HS, as described. 21 The luciferase reporter plasmids, constructed with both alleles, were transiently transfected into the cells using the TransFast lipofection reagent (Promega). The Renilla luciferase expression plasmid, pRL-TK, was co-transfected as an internal standard.…”
