C‐type lectins are one of the pattern‐recognition proteins involved in innate immunity in invertebrates. Although there are 16 C‐type lectin genes that have been identified in the genome of Tribolium castaneum, their functions and mechanisms in innate immunity remain unknown. Here, we identified one C‐type lectin orthologue, TcCTL6 (TC003708), by sequencing random clones from the cDNA library of the coleopteran beetle, T. castaneum. TcCTL6 contains a 654 bp open reading frame encoding a protein of 217 amino acids that includes a single carbohydrate‐recognition domain. The expression of TcCTL6 was significantly induced by Escherichia coli, Staphylococcus aureus and stimulation with carbohydrates, including lipopolysaccharide and peptidoglycan. A binding assay suggested that the recombinant TcCTL6 not only bound to lipopolysaccharide and peptidoglycan but also bound to Gram‐positive (S. aureus, Bacillus subtilis and Bacillus thuringiensis) and Gram‐negative bacteria (E. coli and Pseudomonas aeruginosa) in the presence of calcium ions. Furthermore, when TcCTL6 was knocked down by RNA interference, four antimicrobial peptides (attacin1, attacin2, coleoptericin1 and coleoptericin2) were significantly decreased. These results demonstrate that TcCTL6 plays a vital role in the immune response towards pathogen infection by influencing the expression of antimicrobial peptides and the agglutination of bacteria in the presence of calcium ions in T. castaneum.

In this study, an alpha-tubulin gene (EsTUBA, GenBank: KJ509188) was isolated and identified from the Chinese mitten crab Eriocheir sinensis. The EsTUBA mRNA expression patterns under larval brachyurization development and immune challenge stress by Vibrio parahaemolyticus injection in adult crabs were evaluated. EsTUBA cDNA was 1,695 base pairs (bp) with an open-reading frame of 1,356 bp encoding 451 amino acids (49.95 kDa) and containing GTPase domain and C-terminal Tubulin domain. Sequence alignment showed that EsTUBA shared 95-98% identity and exactly similar structural features with its counterparts reported in other animals. Phylogenetic analysis indicated that EsTUBA clustered into one group together with Tubulins from other crustaceans. Sequence alignment, structure comparison and bioinformatic analyses revealed that EsTUBA is member of the Tubulin family. EsTUBA was fluctuant expressed in all tested larval development stages and various organs, and the levels of EsTUBA mRNA expressed in hepatopancreas, gill and intestine, are significantly expression fluctuated and induced after bacteria V. parahaemolyticus injection, while in heart, is almost consistently expressed at 0, 6, 12, 24, 36 and 48 h post-injection compared to control. The molecular cloning of EsTUBA gene is important for further study on the function of this gene.

Calnexin (Cnx) and calreticulin (Crt), which are important chaperones in the endoplasmic reticulum (ER), participate in the folding and quality control of client proteins. Cnx and Crt identified from Chinese mitten crab (Eriocheir sinensis) are designated as EsCnx and EsCrt, respectively. EsCnx and EsCrt are expressed in the hemocyte, hepatopancrea, gill, and intestine at the mRNA and protein level. Immunofluorescence analysis indicated that EsCnx and EsCRT are located in the ER. Moreover, the mRNA and protein expression levels of EsCnx and EsCrt were altered by challenge with lipopolysaccharides (LPS), peptidoglycans (PGN), Staphyloccocus aureus, and Vibrio parahaemolyticus. Recombinant EsCnx and EsCrt (rEsCnx and rEsCrt, respectively) proteins can bind to various Gram-positive and Gram-negative bacteria, as well as to different polysaccharides (LPS and PGN). rEsCnx and rEsCrt assisted in the clearance of V. parahaemolyticus in vivo, and the clearance efficiency was impaired after silencing of EsCnx and EsCrt. Our results suggest that the two ER proteins are involved in anti-bacterial immunity in E. sinensis.
