Function and transcript analysis of gibberellin-biosynthetic enzymes in wheat

Abstract: The enzymes gibberellin (GA) 20-oxidase and 3-oxidase are major sites of regulation in GA biosynthesis. We have characterised one member of each of the gene families encoding these enzymes that are highly expressed in elongating stems and in developing and germinating grains of wheat and are therefore likely to have prominent developmental roles in these tissues. We mapped the three homoeologues of the GA 20-oxidase gene TaGA20ox1 to chromosomes 5BL, 5DL and 4AL. TaGA20ox1 is expressed mainly in the nodes and … Show more

“…Searching of the maize draft genome (A Golden Path version 2.0) by using rice GA3oxs identified two homologs, named ZmGA3ox1 and ZmGA3ox2. In alignment with related GA3ox proteins from rice OsGA3ox1, OsGA3ox2 (Itoh et al, 2001), pea (Pisum sativum) PsGA3ox1 (Lester et al, 1997;Martin et al, 1997), Arabidopsis AtGA3ox1 (Chiang et al, 1995), barley (Hordeum vulgare) HvGA3ox2 (Spielmeyer et al, 2004), wheat TaGA3ox2 (Appleford et al, 2006), and Brachypodium distachyon BdGA3ox2 are roughly the same size with conserved residues throughout the entire protein sequences (Fig. 3).…”

“…In Fv SOC1-OX plants in the SD strawberry background, several strawberry homologs of GA20ox and GA3ox genes were activated and an opposite trend was found in Fv SOC1-RNAi plants. Since GA-20 oxidation is a rate-limiting step in GA biosynthesis (Appleford et al, 2006;Middleton et al, 2012), twofold to severalfold changes in the expression levels of Fv GA20ox genes in our transgenic lines is likely to affect GA levels and, consequently, growth. In Arabidopsis, SOC1 is known to mediate the effect of GA on flowering (Moon et al, 2003;Searle et al, 2006), but its role in the regulation of GA biosynthesis is not clear.…”

“…In wheat the three homoeologues of the GA20-oxidase gene TaGA20-ox1 (involved in gibberellin biosynthesis, controlling PHST/seed dormancy) have also been mapped to chromosomes 5BL, 5DL and 4AL (Appleford et al, 2006). GA20-oxidase gene (encoding gibberellin 20 oxidase), mapped on the long arm (located in bin 4AL5-0.66-0.80) of barley chromosome 5H (Li et al, 2004) has been considered to be a candidate gene for dormancy and/or pre-harvest sprouting tolerance (PHST).…”
