Fully human recombinant antibodies against EphA2 from a multi-tumor patient immune library suitable for tumor-targeted therapy

Abstract: Enhanced EphA2 expression is observed in a variety of epithelial-derived malignancies and is an important target for anti-tumor therapy. Currently, Therapeutic monoclonal antibodies against immune checkpoints have shown good efficacy for tumor treatment. In this study, we constructed an immune single-chain fragment variable (scFv) library using peripheral blood mononuclear cells (PBMCs) from 200 patients with a variety of malignant tumors. High affinity scFvs against EphA2 can be easily screened from the immun… Show more

“…Our library size was compatible with the size reported to other constructed libraries, which range from 10 6 to 10 10 [ 38 , 39 , 40 ]. However, it is very laborious and time-consuming to build large libraries (>10 10 ) due to both the use of a large number of blood samples [ 41 ] and bacteria transformations [ 42 , 43 ]. Shui et al [ 39 ] built an immune library against liver cancer with blood samples from four patients, obtaining a library of size 1.7 × 10 7 .…”

“…The use of ScFv immune phage-display libraries has already shown good results in the development of human therapeutic recombinant antibodies for tumor-targeted therapy EphA2 [ 29 ], and in the discovery of ScFv against IL1RAP from a human library [ 30 ].…”

“…The recovery was amplified by infecting E. coli TG1 for the second and third rounds of screening. Phage amplification was carried out previously described [ 34 ]. Titers of eluted phages in every round were calculated to confirm the enrichment after screening.…”
