Fucoidan from Saccharina japonica Alleviates Hyperuricemia-Induced Renal Fibrosis through Inhibiting the JAK2/STAT3 Signaling Pathway

Abstract: Fucoidan is a native sulfated polysaccharide mainly isolated from brown seaweed, with diverse pharmacological activities, such as anti-inflammatory and antifibrosis. Hyperuricemia (HUA) is a common metabolic disease worldwide and mainly causes hyperuricemic nephropathy, including chronic kidney disease and end-stage renal fibrosis. The present study investigated the protective function of fucoidan in renal fibrosis and its pharmacological mechanism. The renal fibrotic model was established with the administrat… Show more

“…The resulting images were assessed by light microscope at a magnification of ×400. As previously described, we assessed the degree of renal damage by assigning scores based on the histopathological changes observed in both the tubular and glomerular regions . The areas of collagen deposition of each section were analyzed and scored with Image-Pro Plus 6.0 software (Media Cybernetics, Bethesda, MD, USA).…”

“…Liver and kidney tissues from the experimental mice were collected, washed with saline, weighed, and the renal index was calculated for each group of mice. The renal index was expressed as a percentage of kidney weight over the whole body weight of the each mouse [ 28 ]. A section of the kidney tissues was fixed in 4% paraformaldehyde for pathological analysis, while the remaining tissues were stored at −80 °C for subsequent experiments The experimental protocols were approved by the Animal Experiment Ethics Committee of Kunming University of Science and Technology, and the approval number is PZWH (Dian) K2019-0017).…”

“…Finally, observations were made using an Olympus IX83 microscope (Tokyo, Japan) under 200× conditions. At the same time, the degree of kidney damage was scored using the method of Sun et al [ 28 ]; 0 is normal (no damage), 1 is 25% (mild), 2 is 25−50% (moderate), 3 is 50−75% (severe), and 4 is more than 75% (very severe).…”

“…According to previous descriptions, fucoidan was extracted from Saccharina japonica with an average MW of 251.2 kDa [31]. Briefly, the seaweed was chopped up and stirred overnight in water at a temperature of 70 • C. After centrifugation, the liquid mixture was mixed with calcium chloride.…”
