FTIR Spectroscopy as a Tool to Study Age-Related Changes in Cardiac and Skeletal Muscle of Female C57BL/6J Mice

Abstract: Studying aging is important to further understand the molecular mechanisms underlying this physiological process and, ideally, to identify a panel of aging biomarkers. Animals, in particular mice, are often used in aging studies, since they mimic important features of human aging, age quickly, and are easy to manipulate. The present work describes the use of Fourier Transform Infrared (FTIR) spectroscopy to identify an age-related spectroscopic profile of the cardiac and skeletal muscle tissues of C57BL/6J fem… Show more

“…Peak area at 2868 cm –1 and 2956 cm –1 are associated to ν s (C–H) or ν as (C–H) of methyl (CH 3 ), respectively [ 22 ]. A ratio including these peak area ((2851 cm –1 + 2922 cm –1 )/(2956 cm –1 + 2868 cm –1 )) can be informative of acyl chain length [ 31 ] and, thus, it was also calculated ( Fig. 4 B, C).…”

“…With spectroscopic data, it is virtually impossible to analyze all variables without the use of multivariate analysis. Here, we used PLS-R supervised multivariate analysis in the three spectral regions, using the 2nd derivative spectra and a random intern cross-validation with Kernel algorithm, as previously explained [ 10 ]. In this case, the Y matrix is the cell passage number which was used to produce a correlation plot between spectral profile and passage number.…”

“…The intensity of specific spectral bands from cells at passages 5, 7, 11 and 17 was calculated using two different approaches as in our previous study [ 10 ]. Briefly, the intensity of peaks assigned to CH (3010 cm −1 ), CH 2 (2851 cm −1 and 2922 cm −1 ) CH 3 (2959 cm −1 and 2871 cm −1 ), C=O (1733 cm −1 ), cholesterol esters (1169 cm −1 ) and protein secondary structures, namely β-sheets (1693 cm −1 , 1682 cm −1 and 1628 cm −1 ), were calculated using the peak intensities of 2nd derivative spectra.…”

“…considering tissues such as blood (56), saliva (57-59), urine (60), bone (61,62), pulmonary oedema fluid (63). On the other hand, there are studies presenting the use of FTIR to analyse molecular profiles of muscle tissue in animal models (64)(65)(66) and human muscle (67, 68). Nevertheless, there are only a few studies that have used FTIR spectroscopy to investigate modifications in muscle tissue associated with DM, and even these have been performed in animal models (69)(70)(71).…”
