From water striders to water bugs: the molecular diversity of aquatic Heteroptera (Gerromorpha, Nepomorpha) of Germany based on DNA barcodes

Abstract: With about 5,000 species worldwide, the Heteroptera or true bugs are the most diverse taxon among the hemimetabolous insects in aquatic and semi-aquatic ecosystems. Species may be found in almost every freshwater environment and have very specific habitat requirements, making them excellent bioindicator organisms for water quality. However, a correct determination by morphology is challenging in many species groups due to high morphological variability and polymorphisms within, but low variability between spec… Show more

“…This local sequence database consists of the compiled data which are based on the DNA library with more than 23,000 barcoded German animal species assembled in two major DNA barcoding campaigns: “Barcoding Fauna Bavarica” (BFB, http://www.faunabavarica.de, Haszprunar, 2009) and “German Barcode of Life” project (GBOL, http://www.bolgermany.de, Geiger et al, 2010), with nearly 250,000 vouchers curated at the Zoological State Collection Munich, Germany (http://www.barcoding-zsm.de). Data releases have been published for all major arthropod groups, that is, Coleoptera (Hendrich et al, 2015; Raupach, Hannig, Moriniere, & Hendrich, 2016; Raupach, Hannig, Morinière, & Hendrich, 2018; Rulik et al, 2017), Diptera (Morinière et al, 2019), Ephemeroptera, Plecoptera, and Trichoptera (Morinière et al, 2017), Heteroptera (Havemann et al, 2018; Raupach et al, 2014), Hymenoptera (Schmid‐Egger et al, 2019; Schmidt, Schmid‐Egger, Morinière, Haszprunar, & Hebert, 2015; Schmidt et al, 2017), Lepidoptera (Hausmann, Haszprunar, & Hebert, 2011a; Hausmann, Haszprunar, Segerer, et al, 2011), Neuroptera (Morinière et al, 2014), Orthoptera (Hawlitschek et al, 2018), Araneae and Opiliones (Astrin et al, 2016), and Myriapoda (Spelda, Reip, Oliveira Biener, & Melzer, 2011; Wesener et al, 2015).…”

“…Sequence divergence was estimated using the K2P model with 1000 bootstrap estimates in MEGA7 [57]. This a standard model that has been extensively used in barcoding studies [58,59]. MOTUs or barcode species were delimited using 2% in divergence cut-off, which has proved to be consistent for most invertebrate groups [6062].…”

“…However, the genetic diversity of A. aestivalis has not been investigated in wide-scale studies. Available data have been obtained during complex studies based on DNA barcodes in aquatic Heteroptera (Havemann et al 2018), resolving relationships among related species (Hebsgaard et al 2004;Carbonell et al 2011) or in approaches leading to the effectiveness evaluation of new molecular tools (Sonnenberg et al 2007). Recently, the karyotype and male meiosis in A. aestivalis have been examined (Stoianova et al 2017).…”
