From Small Molecules toward Whole Cells Detection: Application of Electrochemical Aptasensors in Modern Medical Diagnostics

Abstract: This paper focuses on the current state of art as well as on future trends in electrochemical aptasensors application in medical diagnostics. The origin of aptamers is presented along with the description of the process known as SELEX. This is followed by the description of the broad spectrum of aptamer-based sensors for the electrochemical detection of various diagnostically relevant analytes, including metal cations, abused drugs, neurotransmitters, cancer, cardiac and coagulation biomarkers, circulating tum… Show more

“…The structured RNA motifs were termed aptamers based on the Latin words "aptus" (fitting) or "aptare" (to fit) and the Greek word "meros" meaning part. Highaffinity nucleic acid ligands for a protein were isolated by systematic evolution of ligands by exponential enrichment (SELEX) that depends on alternate cycles of ligand selection from pools of variant sequences and amplification of the bound species [2][3][4][5][6][7]. Several rounds of selection exponentially enriched the population for highest affinity species that can be clonally isolated and characterized.…”

“…At present, aptamers are used to construct various types of biosensors, such as fluorescence aptasensors, electrochemical aptasensors and colorimetric aptasensors [ 24 , 25 , 26 ]. Among them, the fluorescence aptasensors have a high sensitivity and the experimental results are more intuitive [ 27 ].…”

“…Numerous electrochemical aptasensors have been developed so far for the detection of single analytes that are important from the points of view of (1) disease diagnostics [ 19 , 20 , 21 , 22 , 23 , 24 ], (2) food analysis [ 25 , 26 , 27 ], and (3) environmental monitoring [ 28 , 29 ]. Moreover, several review papers discussing current achievements and future perspectives in the area of electrochemical aptasensors have recently been presented [ 30 , 31 , 32 ]. Only a few examples are cited here.…”
