From Initial Hit to Crystal Optimization with Microseeding of Human Carbonic Anhydrase IX—A Case Study for Neutron Protein Crystallography

Abstract: Human carbonic anhydrase IX (CA IX) is a multi-domain membrane protein that is therefore difficult to express or crystalize. To prepare crystals that are suitable for neutron studies, we are using only the catalytic domain of CA IX with six surface mutations, named surface variant (SV). The crystallization of CA IX SV, and also partly deuterated CA IX SV, was enabled by the use of microseed matrix screening (MMS). Only three drops with crystals were obtained after initial sparse matrix screening, and these wer… Show more

“…The largest CA IX SV crystal that we obtained was 0.8 mm 3 and efforts to scale up and increase the volume continue (see Fig. 5 in Koruza, Lafumat, Nyblom et al, 2018). We obtained crystals in space group P2 1 with two apparently different unit cells labelled 'small' (unit-cell parameters a = 44.5, b = 65.4, c = 46.7 Å , = 115.1 ) and 'big' (unit-cell parameters a = 48.9, b = 65.1, c = 76.3 Å , = 92.86 ).…”

“…Crystallization drops were prepared using both the hangingdrop and sitting-drop vapour-diffusion methods after a lengthy optimization procedure as described elsewhere (Koruza, Lafumat, Nyblom et al, 2018). Briefly, crystals were initially grown for the preparation of seed stocks using a 1:1 ratio of protein solution (17 mg ml À1 H/H CA IX SV ) and 30%(w/v) PEG 4000, 0.1 M Tris-HCl pH 8.5, 0.2 M sodium acetate or 0.2 M ammonium formate.…”

“…This crystallization phase diagram was deduced from the raw data of Figure S2. This crystallization phase diagram is unique for RuBisCO from A. eutrophus and is not consistent with either the diagram of human carbonic anhydrase IX by Koruza K. et al [24] or nucleation and growth of protein crystals: general principles and assays in Methods in Enzymology [25]. We can understand easily from this canoe-shaped crystallization phase transition diagram both the salt-in effect and the salting out effect simultaneously which are required to crystallize a protein with precipitating agent as salts.…”
