--Effect of pork pancreatic elastase on thickening of the renal glomerular basement membrane was studied in three litter mates of the spontaneously diabetic rats by electron microscopic morphometry. Intramuscular injection of physiologic saline solution or elastase solution (5 mg/kg body weight/day) was repeated daily for 30 days. Renal cortex specimens were obtained before and after the injection in two litter mates. Renal glomerular basement membrane thickness (RGBMT) of the rats injected with saline increased markedly, whereas RGBMT of the rats treated with elastase remained close to the initial value. In each of the three litter mates, RGBMT after the treatment was significantly thinner in the elastase injected rats than in the saline injected rats.---elastase; glomerular basement membrane ; spontaneously diabetic rat Many investigators suggested that the strict control of hyperglycemia could protect the development of diabetic microangiopathy (Miki et al. 1972 ;Tchobroutsky 1978). In addition, hypophysectomy (Ireland et al. 1976) and pancreatic transplantation (Mauer et al. 1975) have also been attempted for this purpose. However, no direct therapy of the microangiopathy has been reported. The diabetic glomerular lesion, one of the microangiopathies, is characterized by thickening of the glomerular basement membrane and the increase in the amount of basement membrane-like matrix within mesangial regions (osterby 1973), and these changes are known to be induced by the accumulation of certain glycoproteins (Spiro 1963). Pancreatic elastase has a collagenolytic action in liver cirrhosis (Fukuhara 1975).
