Dihuang powder (DHP) has been used in the traditional Chinese medicine for the treatment of diarrhea in some regions of China. But up to now, the anti-diarrheal activity of DHP haven’t been performed with modern pharmacological technology. This study aims to investigate the quality control, the potential toxicity and anti-diarrheal activity of Dihuang powder in mice. High performance liquid chromatography (HPLC) and thin layer chromatography (TLC) were used to detect five active compounds in DHP for quality control, and the acute toxicity and sub-acute toxicity for 28-day oral administration of DHP were then evaluated. The anti-diarrheal activity was investigated using mouse model. Results showed that the levels of quercetin and berberine in DHP were 0.054 and 0.632 mg/g, respectively, and atractylodin, matrine, and patehouli aleohal were also detected in DHP. At the given doses, DHP was safe in terms of acute and sub-acute toxicity. Meanwhile, DHP exhibited strong anti-diarrheal effects as well as decreased gastrointestinal motility and the secretions induced by Sennae and castor oil in a dose-dependent manner. It could decrease the content of IL-1β, IL-6, and TNF-α in the small intestine, and improve the histopathological changes of small intestine and large intestine induced by Sennae. The antinociceptive and anti-inflammatory activities in vivo also were presented. Based on all of the results, we thought that DHP has anti-diarrheal activity, and could be used to treat diarrhea as well as alleviate the pain and inflammation induced by diarrhea. This study provides a theoretical basis for the clinical use of DHP and may assist in the development of new drugs for the treatment of diarrhea. The mechanism of the anti-diarrheal activity should be investigated in the future.

Many patients in Africa often utilize herbal medicine for the management of hyperactive gut disorders such as diarrhea and abdominal colic. There is therefore a need to scientifically evaluate efficacy of this herbal remedies. This study investigated the myorelaxant effect of Carissa edulis (Forssk.) Vahl and its possible mechanism of action using isolated rabbit jejunum preparations. Pieces of jejunum were isolated from adult New Zealand white rabbits. They were then mounted in an organ bath containing Tyrode's solution. The rate and force of jejunal contraction were recorded and analyzed using a Powerlab that was coupled to Chart5 software for windows. The effects of the extract (0.1-10.0 mg/ml) on spontaneous contraction were investigated. Its effect (3.0 mg/ml) was also studied in the presence of L G-NAME (nitric oxide synthase inhibitor), methylene blue (soluble guanylyl cyclase inhibitor) and propranolol (non-selective β adrenergic receptors antagonist). The extract dose-dependent significantly decreased the force but not the rate of spontaneous rabbit jejunal contraction. Among the blockers used, only L G-NAME significantly blocked the effect of the extract. Aqueous root bark extracts of C. edulis possess significant myorelaxant effect on isolated rabbit jejunum. This appears to be mediated through stimulation of nitric oxide release by nitric oxide synthase.

Purpose This study aims to report the phytochemical screening and anti-diarrhoea properties of methanol extract of Diodia sarmentosa Swartz (DSS) leave (at 200, 400 and 600 mg/kg) in albino rats. Design/methodology/approach Three types of diarrhoea models (castor oil-induced diarrhoea, castor oil-induced gastrointestinal motility and castor oil-induced enteropooling), as well as an assay for serum and faecal electrolyte concentrations, were used. Acute toxicity study and phytochemical screening of the extract were carried out using standard techniques. Findings The percentage inhibition of diarrhoea by DSS 200, 400 and 600 mg/kg was obtained as 25.7%, 55% and 84.6%, respectively, compared with loperamide that had 87.6% inhibition. DSS dose dependently modulated the number and frequency of defecation, the weight of the faeces of the diarrhoea rats, the distance that was travelled by charcoal meal, mean differences in the full lengths of the intestines and the distance travelled by charcoal meal, weight and volume of intestinal contents, serum and faecal concentrations of potassium, sodium, chloride, magnesium, bicarbonate, iron and zinc relative to the negative control and in a manner akin to loperamide (for the extract at 600 mg/kg). DSS or loperamide attenuated the loss of calcium in the faeces of the rats. DSS possesses anti-diarrhoea properties, which may be attributed to the phytochemicals in it. Finally, the study showed the safety in the usage of DSS. Originality/value DSS exerted its anti-diarrhoea action in castor oil-induced diarrhoea rats, by modulating their intestinal secretory and motile activity in a manner akin to loperamide (for DSS at 600 mg/kg).
