Frankiamide: A Structural Revision to Demethyl (C-11) Cezomycin

Abstract: A compound (née frankiamide) isolated from Frankia was initially structurally elucidated on the basis of NMR and MS. However, X-ray analysis provided a structure that is in discord with this structure and in light of this the structure has now been revised and the compound renamed as demethyl (C-11) cezomycin.

“…To date, the secondary metabolome of Frankia has not been explored beyond two reports concerning the production of anthraquinone- and calcimycin-type antibiotics 149,150. The genome sequences of Frankia now certainly provide a rationale to explore the chemistry of these intriguing microorganisms.…”

“…Quinonoid pigments of presumptive polyketide origin and a derivative of the calcium-binding streptomyceteproduced calcimycin antibiotics, demethyl C-11 cezomycin, were isolated from Frankia strains ORS 020604 and AiPs1, respectively, and their structures determined as early examples of Frankia-based natural products (25,30). The most wellstudied area for Frankia has focused on the production of numerous oxidized and cyclopropanated fatty acids and triterpene hopanoids that differ in content between vegetativegrowth cells and those in N 2 -fixing vesicles (54).…”
