FOXO3 signalling links ATM to the p53 apoptotic pathway following DNA damage

Abstract: DNA damage as a result of environmental stress is recognized by sensor proteins that trigger repair mechanisms, or, if repair is unsuccessful, initiate apoptosis. Defects in DNA damage-induced apoptosis promote genomic instability and tumorigenesis. The protein ataxia-telangiectasia mutated (ATM) is activated by DNA double strand breaks and regulates apoptosis via p53. Here we show that FOXO3 interacts with the ATM-Chk2-p53 complex, augments phosphorylation of the complex and induces the formation of nuclear f… Show more

“…Thus, GP protects N2a cells at multiple steps. On the one hand, up-regulation of Akt leads to an increase of Bcl2 expression [34], which promotes cell survival [35] but on the other hand, down-regulation of FOXO3 [36] and p53 [37,38] results in the promotion of apoptosis. This shows that GP has multiple effects on cell death and survival.…”

“…145 However, the relationship between AKT and DNA damage signaling is complex, and many other targets that are inhibited by AKT actually enhance DNA damage checkpoints and repair. For example, FOXO3A has been shown to directly associate with ATM, p53, and Chk2, enhancing the activation of these proteins, 146,147 and to regulate both the G1 148 and G2/M 149 DNA damage checkpoints. Thus, FOXO3A inactivation through AKT phosphorylation might be expected to impair the DDR.…”
