FOXK2 Transcription Factor and Its Emerging Roles in Cancer

Abstract: Forkhead box (FOX) transcription factors compose a large family of regulators of key biological processes within a cell. FOXK2 is a member of FOX family, whose biological functions remain relatively unexplored, despite its description in the early nineties. More recently, growing evidence has been pointing towards a role of FOXK2 in cancer, which is likely to be context-dependent and tumour-specific. Here, we provide an overview of important aspects concerning the mechanisms of regulation of FOXK2 expression a… Show more

“…For example, Foxk proteins promote the expression of genes encoding components from the glycolytic pathway and hence contribute to metabolic reprogramming ( 15 , 16 ). There is a growing appreciation of the biological importance of the FOXK proteins and their deregulation in diseases such as cancer (reviewed in 17 ). Developmentally, FoxK has been implicated in TGF-β signalling during midgut differentiation in Drosophila ( 18 ) and also has a role in stem cell biology as the mouse homologue of FOXK1, MNF, controls the proliferation of myogenic stem cells ( 8 , 19 ).…”

“…Growing evidence shows that FOXK2 participates in a range of key processes in cancer cells, such as cell proliferation, invasion and migration, DNA damage, and autophagy [46]. The function of FOXK2 is context and tumor-speci c, and it might serve as an oncogene and tumor suppressor in different cancers [47]. It was reported that FOXK2 inhibited cell proliferation and invasion in breast cancer, non-small cell lung cancer (NSCLC), clear-cell renal cell carcinoma, gastric cancer, and indicated favorable prognosis [48][49][50][51].…”

“…5G). Transcription factor (TF) binding to promoters or enhancers is essential for gene transcription, which can activate or repress the expression of genes in a sequencespeci c manner 6 . To further explore this possibility, several TFs were analysed.…”
