The aim of the present study is to evaluate the cupric ion releases and cytotoxicity of Cu-bearing intrauterine device (IUD) with different surface areas. TCu380, GCu300 and MCu165 were selected and extracted with MEM for 24h. Inductively Coupled Plasma Mass Spectrometry (ICP-MS) was employed to determine the concentration of Cu ions. MTT assay was conducted to evaluate the cell viability of original and diluted extracts. Oxidative stress indicators such as superoxide dismutase (SOD) and lipid peroxidation (MDA) were measured. The results showed the concentration of Cu ions released from three kinds of IUDs were 114.836mg/L, 91.161mg/L and 85.750mg/L, respectively. Cell viability of all original IUDs extracts was around 10%. TCu380 showed more significant cytotoxicity and higher oxidative stress level than other two IUDs when extracts were diluted. The study suggested that there was a positive correlation between the quantity of released Cu ions and cell damage effect in Cu-IUDs. The mechanism of cytotoxicity might be elevation of oxidative stress.

We immersed copper T380A and T220C IUDs in simulated uterine solution (SUS) and also in simulated body fluid (SBF). The release rate and corrosion products of copper ions after soaking in SUS and SBF for different time spans were studied by an atomic absorption spectrophotometer, scanning electron microscopy with energy dispersive x-ray microanalysis (SEM/EDX) and x-ray diffraction. The highest corrosion rate of the TCu380A IUD was obtained on the first day, while the highest corrosion rate of the TCu220C IUD was obtained on the third day. The copper corrosion rate of the TCu380A IUD was much faster than that of the TCu220C IUD in the first few days. The corrosion extent of copper wire was higher than that of a copper tube when first immersed in solution. For the same IUD in the same solution, the corrosion extent in SBF was higher than in SUS. The main compounds identified were cuprite (Cu(2)O). Copper wire and copper tubes were implanted in the uteruses of rats. In vivo experiments were also done. The corrosion rate in vivo was higher than that in vitro.

A study was undertaken to look at the clinical behavior of the Dalkon Shield, the Lippes Loop D and the TCu-200 inserted by physicians and midwives. Differences among the three devices were not statistically significant, although lower expulsion rates and higher pregnancy rates were reported for the Dalkon Shield. No differences were observed between insertions performed by physicians and those performed by midwives; this emphasizes the efficiency of using paramedical personnel in the field of intrauterine contraception.
