Four New Species of Amanita in Inje County, Korea

Abstract: Amanita (Agaricales, Basidiomycota) is one of the most well-known genera composed of poisonous mushrooms. This genus of almost 500 species is distributed worldwide. Approximately 240 macrofungi were collected through an ongoing survey of indigenous fungi of Mt. Jeombong in Inje County, Korea in 2014. Among these specimens, 25 were identified as members of Amanita using macroscopic features. Specimens were identified to the species level by microscopic features and molecular sequence analyses of the internal tr… Show more

“…It is di cult to differentiate closely related species. However, molecular techniques such as polymerase chain reaction (PCR) for mushroom identi cation have proved to be fast, speci c, and sensitive, especially, inconsistent morphology and indiscrimination among species of mushrooms (Cho et al 2015;Kim et al 2016;Lee et al 2000;Persoh et al 2009;Raja et al 2017). The conserved region such as Internal Transcribed Spacer (ITS) of nuclear ribosomal DNA (rDNA) usually has been targeted because it is highly conserved among interspecies but variable between intraspecies (Raja et al 2017).…”

“…The genus Amanita consists of 95% of the species which are characterized by agaricoid basidiomata, colorless and hyaline, ballistosporic and smooth basidiospores, free lamellae, presence of volval remnants (Persoon 1797). A total of 540 known species of Amanita are distributed worldwide (Yang 2000, Kirk et al 2008, Menolli et al 2009, Tulloss 2009, Wartchow et al 2009, Justo et al 2010, Wartchow and Gamboa-Trujillo 2012, Cho et al 2015, Hosen et al 2015, Tang et al 2015, Wartchow and Cortez 2016, Jabeen et al 2017, Cui et al 2018, Kiran et al 2018a, b). From Pakistan, 19 species of Amanita are known to date (Ahmad et al 1997, Jabeen et al 2017, Kiran et al 2018a, b).…”

“…Although several coding genes have been used in recent fungal phylogeny studies, the ITS region remains the most commonly used locus for fungal phylogenetics, and the accumulated DNA sequence information for the ITS region has greatly improved the accuracy of identification in the Fungi [ 14 15 16 17 ]. Previously, we showed the efficiency and accuracy of a two-step approach for fungal identification: phylogenetic analysis using ITS sequences followed by confirmation based on morphological observations [ 18 19 20 21 ].…”
