Four‐day continuous gastric pH monitoring following anti‐acid secretory drug administration: cross‐over test to assess the early effects

Abstract: SUMMARY BackgroundThere have been few reports that compare the effect of histamine H2-receptor antagonists (H2RAs) and proton pump inhibitors (PPIs) using continuous gastric pH monitoring for a long duration.

“…The main goal of this study was to test the feasibility of a capsule normally used in the clinical setting in humans to measure gastroesophageal reflux disease (Bravo system) for monitoring intragastric pH in the rat. The Bravo capsule system has primarily been used in humans [11][12][13][14] , but also in animals [15] for diagnosis of gastroesophageal reflux disease.…”

“…First, our new technique had a few early dislodgements of the Bravo capsules with meals. Changing the fixation site of the capsule to a higher position in the stomach in order to avoid strong gastric contractions or using more clips, which has been demonstrated in other studies [15,19], will help solve this problem. Second, the change in distance from the capsule to the catheter on day 2 is most likely due to movement of the flexible catheter tip during the study.…”

“…Several studies have attempted to measure intragastric pH using the Bravo capsule in humans and monkeys [15][16][17][18][19]. Use of the Bravo capsule in humans intragastric pH monitoring, fixed by mucosal suction, has been reported, and the Bravo capsules remained on the gastric wall for over 48 h in 82% of cases [16].…”

“…Furthermore, placebo effect could be higher because we set the baseline symptoms of heartburn only once or more in the observation period in our study. H 2 RAs have been reported to inhibit gastric acid secretion more promptly than PPIs and are therefore expected to rapidly improve symptoms of heartburn [18][19][20][21].…”
