Formulation technologies for oral vaccines

Abstract: Summary Many options now exist for constructing oral vaccines which, in experimental systems, have shown themselves to be able to generate highly effective immunity against infectious diseases. Their suitability for implementation in clinical practice, however, for prevention of outbreaks, particularly in low‐ and middle‐income countries (LMIC), is not always guaranteed, because of factors such as cost, logistics and cultural and environmental conditions. This brief overview provides a summary of the various a… Show more

“…With subunit and recombinant vaccines, protection of the antigen from the harsh stomach environment will need to be addressed, and formulations can be tailored to the target species. [116][117][118][119][120] With distribution in the wild, vaccine formulations will need to be protected from environmental factors such as rain and sunshine.…”

“…Drugs in general are absorbed faster and access directly the blood circulation bypassing the liver; therefore, avoiding hepatic clearance [ 209 ]. The convenience of oral formulation has led to significant efforts in the formulation of oral vaccines [ 210 , 211 , 212 , 213 ]; however, oral mucosa delivery is not generally considered for conventional vaccines due to a limited accessibility and its low immunogenic potential.…”

“…Other parts of the intestine also play a role, particularly the lamina propria, a compartment stretching the length of the intestine basal to the enterocytes, feeding into the intestinal lymphatics, and a population of intra-epithelial lymphocytes (IEL) embedded within the tight junctions of the much larger epithelial cells. For a general review of formulations for oral vaccines, see New [ 4 ].…”
