Background: Fast disintegrating tablet (FDT) of ondansetron has better patient compliance as it is easy to swallow for the treatment of nausea and vomiting. Moreover, fast absorption, rapid onset, reduced loss of drugs, and enhanced bioavailability are making oral dispersible systems a better drug delivery system. This research was conducted with the objective to formulate and evaluate the oral FDT of ondansetron HCl using natural polymers. Methods: The tablets were formulated employing the direct compression technique using different ratios of natural super disintegrants such as spray-dried banana powder, ispaghula husk powder, and combination of both polymers. The developed powders were subjected to the pre-compression parameters and the powders were punched using a single-head rotatory compression machine. The formulated 200 mg tablets were subjected to post-compression evaluation. Results: The Fourier-transform infrared spectroscopy studies showed no chemical interaction between pure drugs, natural super disintegrants, and other excipients. Formulation F6 containing ispaghula husk powder in a 1:4 ratio demonstrated 98.60±0.003% of drug release within 10 minutes indicating the best formulation. Conclusion: FDT of ondansetron was considered the best formulation in the drug delivery system which followed the Higuchi model with the Fickian release.

The study aimed to analyze differences in the effectiveness of ondansetron administration compared to the administration of ondansetron and the combination of Curcuma xanthorriza and Kleinhovia hospita against the incidence of PONV at Dr. Moewardi Hospital. Thirty patients with the physical status of the American Society of Anesthesiologists (ASA) I or II who underwent laparotomy surgery were randomly premedicated by double-blinded method in each group: the control group, namely premedication of 8 mg ondansetron per oral, and the treatment group, namely premedication of capsule combination of ondansetron 8 mg and Curcuma xanthorriza and Kleinhovia hospita 1 gr. PONV was assessed using PONV score assessment. PONV assessment was carried out 2 hours post-operation. PONV scores between the control group and the treatment group was found significant (p = 0.017). There was no significant difference (p > 0.05) in other variables such as age, weight, height, body mass index, and duration of surgery. Risk factors such as gender and smoking history were obtained PONV scores with insignificant differences (consecutive p values = 0.625; 0.527). Premedication of a combination of ondansetron and Curcuma xanthorriza and Kleinhovia hospita in this study proved more effective in reducing PONV scores.

Our goal is to create and validate a UV Spectrophotometric method for estimating the Ondansetron HCL. According to ICH criteria, the procedure was verified. At 248.8nm, ondansetron has its highest absorption. Between 2 and 22g/ml, the linearity was noted. Recovery studies supported the analysis' findings. The findings of the validation parameters showed that the suggested technique was determined to be exact, precise, repeatable, sensitive, and appropriate for regular quality control analysis for estimation of ondansetron hydrochloride in solid dose formulation.
