Formation and Evaluation of a Two-Phase Polymer System in Human Plasma as a Method for Extracellular Nanovesicle Isolation

Abstract: The aim of the study was to explore the polyethylene glycol–dextran two-phase polymer system formed in human plasma to isolate the exosome-enriched fraction of plasma extracellular nanovesicles (ENVs). Systematic analysis was performed to determine the optimal combination of the polymer mixture parameters (molecular mass and concentration) that resulted in phase separation. The separated phases were analyzed by nanoparticle tracking analysis and Raman spectroscopy. The isolated vesicles were characterized by a… Show more

“…Raman spectrum of EVs isolated by DF (Figure S5) agrees with previous reports by Krafft et al 50 and Slyusarenko et al 51 and contains the expected peaks assigned in Table S3. Specifically, the peak near 704 cm -1 corresponds to cholesterol and cholesterol esters.…”

“…The small extracellular nanovesicles (SEVs) were isolated from the PPP using a two-polymer system, previously described in [ 19 ]. The polymers, polyethylene glycol (PEG, 20 kDa), and dextran (DEX, 450–650 kDa) (both from Sigma-Aldrich, St. Louis, MO, USA), were dissolved in PBS at concentrations of 30% and 35%, respectively.…”

“…The total population of nanovesicles was isolated from pellet pure plasma (PPP) samples using a two-phase polymer system [ 13 ]. Unless otherwise specified, SEVs were isolated from 2 mL of PPP and resuspended in 100 μL of PBS.…”

“…The total EV population was isolated from PPP using a two-phase polymer system as previously described [ 13 ], with slight modifications. Briefly, a solution of dextran (450–650 kDa, 1.5%) and polyethylene glycol (35 kDa, 3.5%) (both from Sigma-Aldrich, St. Louis, MO, USA) was prepared in the required volume of PPP.…”
