Selective modulation of TRPC6 ion channels is a promising therapeutic approach for neurodegenerative diseases and depression. A significant advancement showcases the selective activation of TRPC6 through metalated type-B PPAP, termed PPAP53. This success stems from PPAP53’s 1,3-diketone motif facilitating metal coordination. PPAP53 is water-soluble and as potent as hyperforin, the gold standard in this field. In contrast to type-A, type-B PPAPs offer advantages such as gram-scale synthesis, easy derivatization, and long-term stability. Our investigations reveal PPAP53 selectively binding to the C-terminus of TRPC6. Although cryoelectron microscopy has resolved the majority of the TRPC6 structure, the binding site in the C-terminus remained unresolved. To address this issue, we employed state-of-the-art artificial-intelligence-based protein structure prediction algorithms to predict the missing region. Our computational results, validated against experimental data, indicate that PPAP53 binds to the 777LLKL780-region of the C-terminus, thus providing critical insights into the binding mechanism of PPAP53.

Background: Ficus religiosa, also known as the peepal tree, is used to treat multiple diseases. It is proved to be effective in treating cognitive impairment. However, the potential targets and pharmacological and molecular mechanisms of its action on the management of Alzheimer’s Disease (AD) are not entirely clear. Therefore, a network pharmacology approach is required to further study and explore its treatment mechanism. Methods: ChEBI database was used to retrieve the active constituents. AD genes were retrieved from the DisGeNet database. SMILES of phytoconstituents were retrieved from PubChem, traced for the positive drug-likeness score, targets were predicted and regulated proteins were enriched to trace probable modulated proteins, pathways, and GO terms. Cytoscape ver. 3.7.2 was used to construct the active ingredient-target-pathway interaction of F. religiosa and network analysis. Molecular docking was also performed. Results: A total of 7 bioactive out of 25 were predicted to possess a positive drug likeliness score. Also, PIK3CA, PIK3RA, AKT1, MAP2K1 and RAF1 were predicted as key gene targets. The results of the GO analysis demonstrated that the somatodendritic compartment, cytoplasm, plasma membrane, and membrane raft are the main cellular components, its molecular functions are mainly catalytic activity, ion binding, protein kinase activity and, identical protein binding. The biological process is focused on biological quality, response to oxygen-contain-compound sound, and metabolic process. Conclusions: This study holistically illuminated possible pharmacological mechanisms of F. religiosa that might be strongly associated with multi-targeting compounds and act on many AD pathways at the same time.

The quality of poultry meat offered to the consumer depends mainly on the level of hygiene during all stages of its production, storage time, and temperature. This study investigated the effect of refrigerated storage on the microbiological contamination, color, and pH of turkey thigh muscles stored at 1 °C over six days. Microbial growth, including total mesophilic aerobes, presumptive lactic acid bacteria, and Enterobacteriaceae, significantly increased, impacting the meat’s sensory attributes and safety. On the 6th day of meat storage, the content of total mesophilic aerobes, presumptive lactic acid bacteria, and Enterobacteriaceae was 1.82 × 107 CFU/g, 1.00 × 104 CFU/g, and 1.87 × 105 CFU/g, respectively. The stability of color was assessed by quantifying the total heme pigments, comparing myoglobin, oxymyoglobin, and metmyoglobin concentrations, analyzing color parameters L*, a*, b*, and the sensory assessment of surface color, showing a decline in total heme pigments, three myoglobin forms, redness (a*) and lightness (L*). In contrast, yellowness (b*) increased. These changes were correlated with the growth of spoilage microorganisms that influenced the meat’s pigmentation and pH, with a notable rise in pH associated with microbial metabolization. Based on the conducted research, it was found that the maximum storage time of turkey thigh muscles at a temperature of 1 °C is 4 days. On the 4th day of storage, the total mesophilic aerobe content was 3.5 × 105 CFU/g. This study underscores the critical need for maintaining controlled refrigeration conditions to mitigate spoilage, ensuring food safety, and preserving turkey meat’s sensory and nutritional qualities. There is a need for further research to improve turkey meat storage techniques under specific temperature conditions by studying the impact of using varying packaging materials (with different barrier properties) or the application of natural preservatives. Additionally, future studies could focus on evaluating the effectiveness of cold chain management practices to ensure the quality and safety of turkey products during storage. By addressing these research gaps, practitioners and researchers can contribute to developing more efficient and sustainable turkey meat supply chains, which may help mitigate food wastage by safeguarding the quality and safety of the meat.
