Follicle-Stimulating Hormone Peptide Can Facilitate Paclitaxel Nanoparticles to Target Ovarian Carcinoma In vivo

Abstract: Chemotherapy is an important treatment for ovarian cancer. However, conventional chemotherapy has inevitable drawbacks due to side effects from nonspecific biodistribution of the chemotherapeutic drugs. To solve such problem, targeted delivery approaches were developed. The targeted delivery approaches combine drug carriers with the targeting system and can preferentially bring drugs to the targeted sites. Follicle-stimulating hormone receptor (FSHR) is an ovarian cancer-specific receptor. By using a peptide d… Show more

“…Gonadotropins activate the MAPK/ERK1/2 signalling pathway in EOC cells OVCAR-3 cells have previously been shown to express the FSHR (Choi et al 2002, Zhang et al 2009). To confirm the expression of both FSHR and LHR, OV207 and OVCAR-3 cells were subjected to immunofluorescent staining (Supplementary Figure 1A and B, see section on supplementary data given at the end of this article), which clearly indicated the presence of both receptors in both cell types.…”

“…FSH affects various aspects of ovarian cancer metastasis, such as it suppresses apoptosis, supports tumor growth by inducing increased expression of VEGFA (VEGF), facilitates blood vessel growth, and may alter certain oncogenic pathways that facilitate proliferative and invasive phenotype. Indeed, studies using nanoparticles conjugated to the b subunit of FSH showed a lot of promise both in vitro and in vivo to increase the specificity and delivery of chemotherapeutic drugs (Zhang et al 2009). …”

“…Subsequently, 1 mL of each of the reconstituted PTX-SNs and PTX-HSNs (3 mg/mL) was added to the dialysis tubing attached to a support, and the release of PTX was determined according to Apparatus 2 of the Unites States Pharmacopeia (USP) dissolution method (paddle method). The release test was performed with 900 mL of the release medium at 100 rpm and 37°C; 1 mL of the released sample was collected at each time point (1,3,6,12,24,36,48,72,96,120, and 144 h). The content of PTX in each sample was measured using the HPLC method described.…”
