Folic Acid Functionalized PFBT Fluorescent Polymer Dots for Tumor Imaging

Abstract: Folic acid functionalized PFBT-COOH and PFBT-NH 2 polymer dots were prepared using a nano precipitation method, and their applications for tumor imaging in vitro and in vivo were demonstrated for the first time. The synthesized FA-PFBT-COOH polymer dots were 50-60 nm in diameter with a zeta potential of greater than 30 mV in water, while the synthesized FA-PFBT-NH 2 polymer dots were 90-105 nm in diameter with a zeta potential of greater than 20 mV in water. Furthermore, the FA-PFBT-COOH polymer dots produce… Show more

“…The FA-MEH-PPV-COOH, MEH-PPV-COOH, FA-MEH-PPV-NH 2 and MEH-PPV-NH 2 polymer dots were prepared by the coprecipitation according to our previous study, [39][40][41][42][43] with some modications. In a typical procedure, a solution of 2 mL of THF containing 250 mg MEH-PPV and 250 mg PS-PEG-COOH or 250 mg MMA-NH 2 and 5 mg NIR775 with/without 50 mg PE-FA was mixed in stock.…”

“…And NIR emission can be realized based on the uorescence resonance energy transfer (FRET) effect. 30 In this study, NIR775-doped poly [2-methoxy-5-(2-ethylhexyloxy)-1,4-phenylenevinylene] (MEH-PPV) polymer dots 39 were used as NIR uorescence imaging probes for in vitro and in vivo cell tracking macrophage Ana-1 cells. In addition, by combining probe-based confocal laser endomicroscopy (pCLE), real-time imaging macrophage cell in deep organs and lymphatics were obtained.…”

“…For example, in the case of a nanosystem formed by gold nanoparticles and quantum dots, the final emitted fluorescence intensity of which could either be enhanced or quenched (turned “ON” or “OFF”) by altering the distance between the two components, thus realizing real‐time tracing at desired sites or situations . One representative example here is the fluorescent polymer dots poly(9,9‐dioctylfluorene‐2,7‐diyl‐ co ‐benzothiadiazole) (PFBT; Figure S1, Supporting Information), a class of macromolecules with various π‐extended bridges, have emerged as powerful contrast agent due to their high brightness, good quantum yield, excellent photostability, and good biocompatibility . Here, PFBT could be dispersed into aqueous solutions by conjugating itself onto the HGCNs though the AuS bond.…”
