Fluorescently labeled pulmonary surfactant protein C in spread phospholipid monolayers

Abstract: Pulmonary surfactant, a lipid-protein complex, secreted into the fluid lining of lungs prevents alveolar collapse at low lung volumes. Pulmonary surfactant protein C (SP-C), an acylated, hydrophobic, alpha-helical peptide, enhances the surface activity of pulmonary surfactant lipids. Fluorescein-labeled SP-C (F-SP-C) (3, 6, 12 wt%) in dipalmitoylphosphatidylcholine (DPPC), and DPPC:dipalmitoylphosphatidylglycerol (DPPG) [DPPC:DPPG 7:3 mol/mol] in spread monolayers was studied by epifluorescence microscopy. “…The difference might be explained by an effect of SP-B and SP-C in Curosurf1. This hypothesis is supported by the experiments of NAG et al [19] who showed that the addition of SP-C to phospholipids modifies the transition of LE-to LC-phases. Enigmatic, however, are the differences in surface topography between porcine lipid extract (Curosurf1) films containing SP-B and SP-C on the one hand, and bovine lipid extract surfactant films with different concentrations of SP-B and SP-C.…”

“…Derivatization of amine groups in SP-C requires treatment of the protein at mild alkaline pH, to ensure deprotonation and reactivity of the amine functions (21)(22)(23)(24). On the other hand, incubation of the protein at basic pH may produce cleavage of the labile thioesther bonds that attach palmitic chains to the two cysteines in the N-terminal segment of the protein (2, 13).…”

“…In the past we used amine-reactive reagents to introduce different groups in SP-C, such as visible-emission fluorescent (22,23) or spin (24) probes. Derivatization of amines is critically dependent on pH as they are good nucleophiles only in their nonprotonated state.…”
