Fluorescence microscopy-based quantitation of GLUT4 translocation

Abstract: Postprandial insulin-stimulated glucose uptake into target tissue is crucial for the maintenance of normal blood glucose homeostasis. This step is rate-limited by the number of facilitative glucose transporters type 4 (GLUT4) present in the plasma membrane. Since insulin resistance and impaired GLUT4 translocation are associated with the development of metabolic disorders such as type 2 diabetes, this transporter has become an important target of antidiabetic drug research. The application of screening approac… Show more

“…The primary antibody (1° Ab) GLUT-4 was diluted in 3% BSA with 0.1% Triton-X (1 : 500), and the attached cells were incubated with 1° Ab overnight at 4 °C. After washing with PBS, the cells were further incubated with the secondary antibody (2° Ab) mouse Alexa fluor (1 : 1000) at room temperature for 1 h. 24…”
