The high sensitivity of optical detection techniques and the highly specific reactions between antibodies and antigens mean that optical immunoassays have attracted much interest in the fields of protein, hormone, drug, and microorganism detection, without the need for complex separation and extraction steps. The immobilization of an antibody on a solid support is a crucial step for optical immunoassays. This review surveys the latest advances in current antibody immobilization techniques in detail, including physical adsorption, covalent attachment, bioaffinity immobilization, and some recently developed methods. Furthermore, specific consideration is given to oriented immobilization, which may improve the homogeneous surface covering the accessibility of the active site and surface coverage, and the analytical performance of immunoassays. Finally, new perspectives for antibody immobilization techniques in optical immunoassays are outlined.

A fiber-optic fluoroimmunoassay system for Dermatophagoides farinae allergen (Der f1) is developed and applied to a flow analysis system. The immunoassay system consists of an optical fiber probe with a collective lens, a reaction cell with inlet/outlet ports for reagents, a laser diode for excitation light source, and a photodiode for detecting fluorescence. The measurement principle is based on sandwich immunoassay. By flowing reagents into the reaction cell, the optical fiber probe becomes coated with immunological complexes formed by capture antibodies, Der f1 derived from house dust mite D. farinae as target analytes, and fluorescent dye (cyanine 5)-labeled antibodies. An excitation light is transmitted into the optical fiber probe, and then the fluorescent molecules of fluorophore-labeled antibodies are excited by the evanescent light of the laser diode. The fluorescence recoupled into the probe is quantified by the photodiode as current values. The calibration range for Der f1 is from 0.98 to 250 ng/ml, and the assays are completed within 16 min. To achieve faster immunoassay, changes in fluorescence signal are monitored using the probe exposed to the fluorescent-labeled antibodies for the detection of Der f1, and calculated to obtain the rate of fluorescence increase for each Der f1 concentration. The calibration range is equal to that of the above-mentioned assay, and the assay time is shortened to 6 min. In the future, the flow immunoassay system allows determining allergens with high precision in a residential environment on site.

A fiber-optic fluoroimmunoassay system for house dust mite allergen (Der f1) is proposed. We optimized or selected optical components for a system, such as optical filters, fluorescence detectors, and optical fiber probes, to detect fluorescence of a cyanine dye (Cy5). We constructed the excitation-emission system for optimized conditions and measured fluorescence intensitites of dilute solutions of Cy5. The measurement range was from 10 0 to 10 5 nmol/l. Next we applied the optical system to fluoroimmunoassays for Der f1. Anti-Der f1 capture antibodies were immobilized onto the end surface of a probe by physical adsorption. Diluted Der f1 solutions and Cy5-labeled antibodies were incubated with capture antibodies to form immune compexes. After all immune reactions were finished, fluorescence from antibodies was measured using the system. Der f1 was quantified in the range of 31.25 to 500 ng/ml. This study suggests that a fluoroimmunoassay system is a base technology to construct a monitoring device for enviromental allergens.
