Fluorescence imaging for photodynamic therapy of non-melanoma skin malignancies – A retrospective clinical study

Abstract: Photodynamic therapy (PDT) has been successfully used in clinical practice for decades; however, clinical outcome data are not always consistent resulting in a great necessity for real-time monitoring to predict the therapy outcome. In a retrospective clinical study, 402 patients with non-melanoma skin malignancies were enrolled who underwent PDT treatment and fluorescence real-time imaging. The photosensitizer used was a chlorine e6 derivative (FotoditazinBaseline

“…Silica gel of 60 0.05-0.20 mm (MachereyNagel) was used for column chromatography. 10,17,12,19,k,p] (1,5,9,13)tetraazacyclohexadecene (1) was obtained by the reaction of 0.26 g (0.91 mmol) of 5-amino-2-dodecyl-3-imino-1,2,4-thiadiazoline (2), synthesized by method described in the literature, [6] and 0.4 g (2.74 mmol) of 1,3-diiminoisoindoline in 3 mL of phenol with step-by-step heating during 1.5-2 hours. The reaction mixture was stirred for 50 hours at temperature 100-110 °С, then the product was washed by hot water and hexane in Soxlet apparatus while absence of phenol, which was determined by qualitative reaction with ferric chloride.…”

“…The achieved PE values confirm the efficiency of the performed procedure, since the level of 40% is typically considered as a success. 28 Averaged photobleaching efficiencies as the function of the delivered light dose at the wavelength of 405 and 660 nm are shown in Figs. 3(c) and 3(d), respectively.…”

“…Two specific absorption peaks in combination with fluorescent properties of chlorin e6 PS provide additional diagnostic opportunities for fluorescence imaging (FI). FI is widely employed for monitoring the PS accumulation and distribution in biotissue prior to a PDT procedure 1,[27][28][29] and for evaluation of PS photobleaching in the course of the procedure. 30,31 Fluorescence excitation at two different wavelengths from the red and blue spectral ranges provides the opportunity to detect fluorescence response of PS from different depths depending on the penetration of the probing radiation into the tissue.…”
