Paracetamol is an extensive analgesic and antipyretic drug. Several batch methods have been reported for the determination of paracetamol in pharmaceutical preparations, such as spectrophotometry, 1-6 reflectance near-infrared spectroscopy, 7 chemiluminescence 8 and liquid chromatography. 2 A number of flow-injection (FI) methods have also been reported for the determination of paracetamol, such as FI-spectrophotometry, using different on-line derivatization reactions. However, the control of such reactions and/or manifolds is still complicated. 9-12 Some methods, such as FI-FTIR 13 and FI with a boron-doped diamond thin film electrode, 14 involve relatively higher cost instruments.A further generation of FIA, SIA is a great potential technique for chemical analysis due to much less reagent consumption, its simplicity and convenience with which manipulation can be automated.So far, an SI procedure for paracetamol determination has been reported. It is based on a reaction with hexacyanoferrate(III), followed by a reaction with phenol at elevated temperature in aqueous ammonia. 15 Simple and inexpensive spectrophotometric flow-injection and sequential-injection systems for the determination of paracetamol using simple reagents based on preliminary concepts introduced at the ICFIA 2001 Conference, 16 are presented. The procedures employ the reaction of paracetamol with nitrous acid at room temperature, producing a derivative. Experimental Chemicals and reagentsAll of the reagents used were of analytical reagent grade. Deionized water was used throughout the experiments. A stock solution (5000 mg/l) of 4-acetaminophenol standard (paracetamol, Fluka, Switzerland), which was assayed using the USP method, 18 was prepared by dissolving 0.5146 g of the standard in water and diluting to the mark in a 100 ml volumetric flask. Working standards were freshly prepared by diluting the stock solution with water to obtain appropriate concentrations. Sample preparationSome locally commercial pharmaceutical preparations were taken as samples to be assayed. For tablet samples, 20 tablets were accurately weighed and finely powdered. An amount of the powder equivalent to a tablet was dissolved in water. After stirring for 15 min at a temperature of 60 -70˚C, the volume was made to 250 ml. The solution was then filtered before analysis. Further appropriate dilutions for FIA procedures were made using water.For a syrup sample, an aliquot corresponding to 200 mg of paracetamol was diluted to 100 ml

KEYWORDSAccurate, precise and simple spectrophotometric methods have been developed and validated for the determination of paracetamol (PAR) and dantrolene sodium (DAS). Spectrophotometric methods including zero order, first derivative ( 1 D) and derivative ratio methods ( 1 DD) have been developed. The zero order spectrophotometric method was used for the determination of DAS in the range of 1-20 μg/mL by measuring the absorbance at 379 nm where PAR exhibits zero reading. 1 D and 1 DD methods were used for the determination of PAR in the range of 1.5-20.0 μg/mL by measuring the peaks amplitudes at 265.5 nm and 265.0 nm, respectively. The proposed methods were used to determine PAR in binary mixture with DAS in the laboratory prepared mixtures and in pharmaceutical dosage form. The results obtained were statistically evaluated and found to be accurate and precise and can be satisfactorily applied for the quality control analysis of the cited drugs.

Resumo: O uso do ácido nitroso para determinação de paracetamol (acetoaminofenol) em formulações sólidas e líquidas empregando um sistema de análise por injeção em fluxo é descrito. A determinação da droga foi realizada pela reação do paracetamol com ácido nitroso gerado em linha e o produto dessa reação foi monitorado em 430 nm em meio alcalino. A curva analítica apresentou uma faixa de concentração de paracetamol variando entre 9,7x10 -5 a 6,2x10 -3 mol L -1 com um limite de detecção de 2,5x10 -5 mol L -1. Os desvios padrão relativos foram menores que 2% para soluções contendo 1,2x10-4 e 2,8x10 -4 mol L -1 de paracetamol (n=10) e uma freqüência de amostragem de 60 determinações por hora foram obtidos. Os resultados da determinação de paracetamol nas formulações farmacêuticas obtidas pelo procedimento em fluxo proposto estão em boa concordância com os resultados obtidos utilizando-se o procedimento da Farmacopéia Americana a um nível de confiança de 95% e dentro de uma faixa aceitável de erro.Palavras-chave: análise em fluxo, paracetamol, formulações farmacêuticas. IntroduçãoParacetamol (acetoaminofenol) é um analgé-sico antipirético derivado da fenacetina, que pode ser administrado associado com outras substâncias, como cafeína [1]. É o analgésico mais vendido no país [2]. Seu uso inadequado pode causar lesões nas células hepáticas, sendo evitado seu uso em casos de dengue, principalmente na forma hemorrágica [3] Uma particularidade dos sistemas de análi-ses de injeção em fluxo (FIA) que contribuiu para sua propagação como técnica analítica é a repetibilidade das operações envolvidas no sistema, como injeção, propulsão das soluções e a regulação tempo dessas operações. Com isso, sistemas de análises por injeção em fluxo permitem que sejam realizadas análises químicas, sem que a reação química tenha se completado, o que é primordial para metodologias utilizadas em bateladas. Considerando-se essas características dos sistemas em fluxo, pode-se empregar reagentes instáveis em diversas determinações analíticas como boa precisão e exatidão, uma vez que a velocidade de decomposição do reagente pode ser controlada. O tempo de residência de um reagente no sistema de análise por injeção em fluxo é da ordem de 1 minuto, assim, o tempo de vida em que um reagente necessita ser estável não deve ser muito superior a este tempo [4,5].Sistemas de análise por injeção em fluxo possibilitam a geração desses reagentes partindose de uma solução com maior estabilidade. No emprego de fortes agentes oxidantes, como Ag(II) e redutores, como U(III), que dificilmente são aplicados em metodologias volumétricas, pode-se partir de soluções de nitrato de prata e acetato de urânio(IV), que são convertidos pela reação com uma substância química (reagentes de Jones, para o agente redutor) ou pela reação eletroquímica (eletrodo de ouro mantido a +2 V vs ECS), para o agente oxidante [5].
