IBrain functions can be affected by the dietary precursors of neurotransmitters. Tryptophan (TRP) is the precursor of serotonin (5-Hydroxytryptamine; 5-HT) neurotransmitter that plays an important role in sleeping, mood, eating behavior, consciousness and cognitive functions. Present study describes the estimation of tryptophan in rat brain by aqueous-reverse-phase high performance liquid chromatography. Estimation was carried out on a shim pack end-capped C-18 column using water as mobile phase (pH=9 adjusted by addition of 0.1 mM NaOH) at a flow rate of 1 ml min-1. Eluents were monitored at 273 nm by an ultra-violet detector. The method was linear (R2=0.9973) and the limit of detection (LOD) and limit of quantification (LOQ) were 4.3920 and 13.3091 ng ml-1, respectively. The developed HPLC method is simple convenient, reliable and economical for the estimation of tryptophan in rat brain. In the present study, Albino Wistar male rats were randomly divided into two groups control and seaweeds treated rats. Seaweeds (Jolyna Laminiriodes) were orally given to test rats at a dose of 200 mg kg-1 whereas control rats were given vehicle for 28 days. After 28 days of treatment, rats were decapitated and its brain samples were taken out for the estimation of TRP. Current study observed increased in TRP levels in the brain samples of seaweed treated rats as compared to control rats. Therefore present study suggested the use of seaweeds in the treatment of disorders caused by TRP depletion. By this newly developed method for the estimation of TRP, effect of various drugs on TRP levels in the brain could be studied.

Chemiluminescence (CL) studies were carried out with luminescent lanthanide ions as probes to a tryptophan-oxidation reaction at pH∼6. The redox system consisted of tryptophan, hydrogen peroxide, and Fe(II) ions (catalysts ofH2O2decomposition). The luminescent lanthanide(III) ions used were Eu(III), Tb(III), Gd(III), and Dy(III). In the case of the reaction system with the Tb(III) ion a significant increase in the chemiluminescence intensity and its duration was observed over the other Ln(III) ions. The CL spectrum registered for this system shows emission bands typical of Tb(III) ions with maxima atλ∼490 and 550 nm, corresponding to the electronic transitions of5D4→7F6and5D4→7F5, respectively. The presence of emission bands characteristic of the Ln(III) ions was also observed in the systems containing Eu(III) and Dy(III) ions. These studies revealed a strong influence of the chemiluminescence intensity associated with the tryptophan oxidation, on the concentration of Ln(III) ions. On the basis of the results obtained, a possible mechanism is proposed for reaction of the systemml: Ln(III)—tryptophan—H2O2–Fe(II), taking into consideration an energy transfer process from the tryptophan oxidation products to the Tb(III), Dy(III), or Eu(III) ions.

A novel galangin-potassium permanganate (KMnO4)-polyphosphoric acid (PPA) system was found to have an outstanding response to tryptophan (Trp). Trp determination using this KMnO4 -PPA system was enhanced significantly in the presence of galangin. A highly sensitive flow-injection chemiluminescence (CL) method to determine Trp was developed based on the CL reaction of galangin-KMnO4 -Trp in PPA media. The presence of galangin, a member of the flavonol class of flavonoid complexes, greatly increased the luminous intensity of Trp in KMnO4 -PPA systems. Under optimized conditions, Trp was determined in the 0.05-10 µg/mL range, with a detection limit (3σ) of 5.0 × 10(-3) µg/mL. The relative standard deviation (RSD) was 1.0% for 11 replicate determinations of 1.0 µg/mL Trp. Two synthetic samples were determined selectively with recoveries of 98.4-100.1% in the presence of other amino acids. The possible mechanism is summarized as follows: excited states of Mn(II)(*) and Mn(III(*) types are the main means of generating chemical luminescent species, and Trp concentration and luminescence intensity have a linear relationship, which enables quantitative analysis.
