Flow cytometry for assessment of the tumor microenvironment in pediatric Hodgkin lymphoma

Abstract: This study indicates that flow cytometry can be used to examine the tumor microenvironment in HL and that percentage of HLA-DR/CD38 copositive lymphocytes may be a biomarker for relapse and refractoriness in pediatric HL.

“…Interestingly, EBV+ cases with Th1 reaction did not show a better prognosis. In contrast, EBV+ pediatric patients exhibited a cytotoxic profile characterized by increased infiltration of CD8 + T cells and showed a better therapeutic response 8,73‐75 . However, a subset of EBV+ patients showed worse outcome with a high number of FOXP3+ cells, suggesting EBV modulates the immune escape strategies by enrichment of the TME with T reg cells and associated, immunosuppressive cytokines 76 .…”

“…There was no correlation between the proportion of BM CD38/HLA-DR co-positive lymphocytes and stage or the presence of B symptoms. CD38/HLA-DR co-positive lymphocyte proportion inversely correlated with CD4/CD8 ratio in the BM as it was observed in the LN tissue 2. There were no significant differences between PB and BM flow cytometric profiles of lymphocyte, monocyte and myeloid cells overall (table 3).…”

“…An independent significant positive prognostic lymphocyte population, characterised by CD38/HLA-DR co-positive cells was lower in the BM compared with the LN tissue 2. Since LN tissue has much higher CD4-positive T cell presence, CD38/HLA-DR co-positive activated CD4-positive cells might play a different role than the same cells present in the BM.…”
