Flow Cytometry: An Introduction

Abstract: A flow cytometer is an instrument that illuminates cells (or other particles) as they flow individually in front of a light source and then detects and correlates the signals from those cells that result from the illumination. In this chapter, each of the aspects of that definition will be described: the characteristics of cells suitable for flow cytometry, methods to illuminate cells, the use of fluidics to guide the cells individually past the illuminating beam, the types of signals emitted by the cells and … Show more

“…By passing a laser beam, the type of event is identified and the droplets containing the events of interest become charged electrostatically. The droplets are then separated by their electrostatic charges in a high voltage field and collected in separate tubes, 96-well plates or slides (144,146,147). However, the jet-in-air cell presentation provides less optical sensitivity than cuvette systems and may negatively influence the identification of extremely rare target cells (119).…”

“…CECs were evaluated using gate for mononuclear cells and data were expressed as cells/mL of blood ( Figure 1). 16,17 The mean, median, maximal, minimal, and standard deviation of values was calculated. The significance of differences between categorical variables were determined by Pearson's Chi-square test or Chisquare with Fisher's exact test.…”
