Flow cytometric DNA analysis as a diagnostic aid for cervical condyloma and cancer

Abstract: Flow cytometric DNA analysis data (FCDA) were obtained from 324 samples provided through the Gynecology‐Oncology Clinic. These samples consisted of 294 combined endoectocervical and vaginal smears and 30 peritoneal washings. Using a conventional scheme for G0/G1, S + G2/M and the coefficient of variation with computer correction for the cell‐cycle kinetics, it was possible to assign a diagnostic Class, I, II, III or V similar to that used by the Cytology Laboratory. These data were then compared with the histo… Show more

“…Histograms were plotted with a Hewlett Packard 7010B X-Y recorder (King of Prussia, PA). DNA histograms were corrected for peak deviations and CV (coefficient of variation) with a program written by the late Dr Walter Tsou (University of Pennsylvania-Presbyterian Medical Center) for an Apple computer (Tsou et a!, 1984).…”

“…The Percoll gradient-purified trophoblasts were fixed in 70% ethanol, stained with 4',6-diamidino-2-phenylindole, and analyzed with an impulse cytophotometer (ICP-22A, Ortho Instruments, Westwood, MA), as described by Tsou et al (18).…”

“…A number of such procedures have been developed using both flow and microscope based image analysis systems. These include slit-scan nuclear cytoplasmic ratios (Wheeless et al, 1984), measurement of DNA content (Tsou et al, 1984;Fujii et al, 1984;Barrett et al, 1979;Tucker, 1979;Sprenger & Witte, 1979), DNA content plus total protein (Linden et al, 1979) and image analysis assaying DNA and chromatin 'texture' (Al & Ploem, 1979;Smeulders et al, 1979). These methods are based on either morphology or non-specific biochemical markers (DNA and total protein).…”
