Flow Cytometric Analysis of Micronuclei in Peripheral Blood Reticulocytes: I. Intra- and Interlaboratory Comparison with Microscopic Scoring

Abstract: Accumulating evidence suggests that reticulocytes (RETs) in the peripheral blood of rats may represent a suitable cell population for use in the micronucleus assay, despite the ability of the rat spleen to selectively remove micronucleated erythrocytes from the peripheral circulation. To evaluate the analytical performance of a previously described flow cytometric method (Torous et al., 2003, Toxicol. Sci. 74, 309-314) that may allow this assay to be conducted using peripheral blood in lieu of bone marrow samp… Show more

“…The erythrocyte suspension (1 Â 10 4 cells per microliter) was incubated with saturating concentrations of monoclonal anti-human CD71 antibody conjugated with phycoerythrin (DB Pharmingeni) and Vybrant Dyecycle Green (Invitrogen) in the dark for 30 min at 37-C and washed with HBSS twice. Finally, the count of CD71 + reticulocytes obtained from 50,000 erythrocytes was measured using a twocolor FACScan flow cytometer (Becton Dickinson) (8,30,31).…”

“…The precision of flow cytometic data acquired by the method employed herein, as well as other analytical performance characteristics of the technique, have been described in detail previously (36)(37). Statistical analyses were performed with JMP Software (v5, SAS Institute, Cary, NC).…”

“…Certainly, fl ow cytometry has been employed for micronucleus analysis with some success and is gaining in popularity [23] . Automated chromosome analysis systems are less reliable and very much in the development phase.…”
