Flexible Synthesis Scheme and Application of AuNP Surface-Conjugatable Metaiodobenzylguanidine Derivatives for Enhanced Cellular Internalization

Natalie S. Potter,
Alan McLean,
Evan C. Bornowski
et al.

Abstract: A synthesis of two metaiodobenzylguanidine (MIBG) derivatives is reported:The design of these molecules allows for their conjugation to a wide range of nanocarriers, such as pulsedlaser ablation (PLA)-synthesized gold nanoparticles (AuNPs), which are utilized here to assess uptake into neuroblastoma (NB), HeLa, and HEK cell lines. Through dark-field imaging and quantitative inductively coupled plasma mass spectrometry (qICP-MS), the MIBG derivatives are found to enhance total AuNP uptake in vitro by over 100× … Show more

“…The only exception that we have reported is in the instance of excess MIBG, which triggers a nonspecific enhanced uptake. 84 Note that this is not the case for this nanosystem. NPs with a 1 μM equivalence of Ce6 were used for the in vitro 2p-PDT experiment, because of their higher dark cell viability over 24 h (Fig.…”

“…49,50 Current work in our group has led to the successful synthesis of a form of MIBG that contains the requisite thiol functional group necessary to be conjugated to 8-arm PEG via the MAL-PEG-SCM linker. 51,52 Preliminary studies have evaluated 8-arm PEG NP photodynamic effects in diseased cells when co-conjugated with photosensitizers such as Ce6. 52 Verteporfin could be another promising photosensitizer since it should be readily conjugatable to the 8-arm PEG NP and is already used for AMD treatment.…”
