Flexible DNA: Genetically Unstable CTG·CAG and CGG·CCG from Human Hereditary Neuromuscular Disease Genes

Abstract: The properties of duplex CTG⅐CAG and CGG⅐CCG, which are involved in the etiology of several hereditary neurodegenerative diseases, were investigated by a variety of methods, including circularization kinetics, apparent helical repeat determination, and polyacrylamide gel electrophoresis. The bending moduli were 1.13 ؋ 10 ؊19 erg⅐cm for CTG and 1.27 ؋ 10 ؊19 erg⅐cm for CGG, ϳ40% less than for random B-DNA. Also, the persistence lengths of the triplet repeat sequences were ϳ60% the value for random B-DNA. Howeve… Show more

“…Both the variance of writhe and the variance of twist may be calculated by statistical mechanics (8) from the length of the Kuhn segment, Ϫ1 (which is twice the persistence length, P), and Poisson's ratio, . Ϫ1 and have been determined experimentally by the kinetics of circularization of restriction fragments containing (CTG) n , (CGG) n , or B-DNA (7,9,10). Fig.…”

“…Thus, the ratios of (CTG) n and (CGG) n are greater than for B-DNA. Since the estimated torsional modulus ␤ for (CTG) n and (CGG) n is within the range of 2.0 -2.4 ϫ 10 Ϫ19 erg⅐cm found for B-DNA by circularization kinetics (7,10,14,15), we conclude that segments of (CTG) n or (CGG) n supercoil more efficiently than B-DNA due to an increased contribution from writhe.…”

“…The plasmid was 2705 bp in length with a total of 338 CG steps (on average, 1 every 8 bp). (CGG⅐CCG) 73 was an XbaI-BamHI restriction fragment from plasmid pRW3691 and contained a total of 156 CG steps (including 10 from the triplet repeat flanking sequences) (47). d(I-C⅐I-C) ϳ7000 (48) give a stock solution of 500 M CG steps.…”

“…(Promega) and were described in detail (12,33,34). The repeat tracts in pRW3546 (12), pRW3691 (34), and pRW3246 (33) are located downstream of the functional lacZ promoter, thus allowing for the direct control of the transcription level by isopropyl-␤-D-thiogalactopyranoside (IPTG) induction and lacI Q repressor inhibition.…”
