Flavonoid Profiles of Two New Approved Romanian Ocimum Hybrids

Abstract: Basil (Ocimum spp.) is a traditional herbal medicine abundant in antioxidants such as phenolic compounds. As part of a diet, this herb is proved to have some roles in decreasing the risk of cancer, and in the treatment of inflammation and neurodegenerative diseases. This study aims to explore the total phenolic and flavonoid content of two new basil hybrids growing in Romania, namely “Aromat de Buzau” (AB) and “Macedon” (MB). The antioxidant capacity of those two species was also analyzed by DPPH and cyclic vo… Show more

“…The determination of the total polyphenol content was performed according to a previously published method, adapted to microplate reader with 96-well plates (Tecan Pro 200, Tecan Trading AG, Männedorf, Switzerland) using Folin-Ciocalteu reagent [41,42]. The Folin-Ciocalteu reagent is a mixture of phosphomolybdic acid (H 3 PMo 12 O 40 ) and phosphotungstic acid (H 3 PW 12 O 40 ) that reacts with phenols and non-phenolic reducing substances to form chromogens which can be detected spectrophotometrically, since in alkaline conditions the oxotungstate and oxomolybdate formed in this redox reaction show a blue coloration proportional to the concentration of polyphenols [43,44]. Briefly, in this method 10 µL of sample were incubated with 25 µL of Folin-Ciocalteu reagent for 5 min, then 25 µL of 20% Na 2 CO 3 and 140 µL of ultrapure water were added and incubated for 30 min at room temperature.…”

“…After 30 min of standing at room temperature, the samples were placed in a Tecan Pro 200 multiwell plate reader (Tecan, Männedorf, Switzerland) and their absorbance was recorded at 760 nm. As the standard references, gallic acid (0.97-500 µg/mL) and tannic acid (0.97-500 µg/mL) were used, and the results were expressed as equivalents of gallic acid (mg GAEq/g) and tannic acid (mg TAEq/g) per 1 g of sample [97][98][99].…”

“…Over time, different methods have been developed for allantoin dosing: alkaline titration, TLC [29], HPLC or spectrophotometry [5,30]. Nowadays, there is a special interest for the discovery of new antioxidant bioactive compounds including polyphenols [31][32][33][34], polysaccharides [35,36], but also naturally derived peptides [37]. Recently, the antioxidant activity was analyzed on different samples obtained from comfrey roots [5,22,26] and snail mucus [4,17,18].…”
