Flavobacterium baculatum sp. nov., a carotenoid and flexirubin-type pigment producing species isolated from flooded paddy field

Abstract: A Gram-stain-negative, aerobic, asporogenous, motile by gliding, dull-yellow, long rod-shaped bacterial strain, designated SNL9T, was isolated from a flooded paddy field near Dongguk University, Republic of Korea. The results of phylogenetic analyses based on 16S rRNA gene sequences indicated that SNL9T represents a member of the genus Flavobacterium and is most closely related to … Show more

“…To test the growth of sand1-3 T cells on different media, the strain was inoculated on R2A agar, nutrient agar (Difco), Luria–Bertani agar (Difco), tryptic soy agar (Difco), marine agar (MB cell) and MacConkey agar (Difco) for 5 days at 30 °C. The pH range was assessed by cultivating cells in R2A broth adjusted to pH 4.0–10.0 (at 1.0 pH unit intervals) using citrate/NaH 2 PO 4 buffer (pH 4.0–5.0), phosphate buffer (pH 6.0–8.0) and Tris buffer (pH 9.0–10.0) at 30 °C for 7 days [47], and NaCl tolerance was tested on R2A agar containing various amounts of NaCl (0–0.5 % in 0.25 % intervals and 1–4 % in 1 % intervals, w/v). Growth of strain sand1-3 T was monitored for 7 days on R2A agar under different temperatures (4, 7, 10, 15, 20, 25, 28, 30, 35, 37, 40, 42, 45, 50 and 55 °C).…”

“…After air-drying, the samples were sliced into small pieces and macerated using a sterile pestle and mortar in sterile distilled water. The macerated samples were serially diluted using 0.85 % NaCl as described previously [18]. The spread plate technique was used for the isolation of novel isolate and the bacterial isolation was performed using Reasoner's 2A (R2A) agar (Difco) at 28 °C for 1 week.…”

“…The presence of exirubin-type pigments were investigated with 20% (w/v) KOH solution ). Hydrolysis of chitin, CM cellulose, starch, and casein was determined as previously described The extraction of cells for carotenoid analysis was performed using a 10 ml methanol/acetone mixture (1:1, v/v) and the absorption spectrum of the pigments was assessed with a spectrophotometer (Multiskan GO, thermos Fisher Scienti c) (Chhetri et al 2021).…”
