Five human tumour cell lines derived from a primary squamous carcinoma of the tongue, two subsequent local recurrences and two nodal metastases

Abstract: Summary.-Five tumour cell lines have been derived from a primary squamous carcinoma of the tongue, from 2 subsequent local recurrences, and from 2 lymph -node metastases-all from the same patient. While the cell lines shared many morphological and biochemical characteristics, those derived from recurrences and metastases appeared to be less differentiated, were less well organized in culture, and displayed fewer desmosomes and tonofilaments than cells in the primary tumour line. A recurrent line showing greate… Show more

“…The derivation and routine culture of cell line HN-5 has been described elsewhere together with its establishment as a transplantable xenograft in immunosuppressed mice. 1,52, 60 Primary epidermal keratinocyte cultures were prepared from skin derived from mastectomies and reduction mammoplasties. Adipose tissue was first dissected away and small (8 mm) cubes of skin were incubated in DMEM containing 0.025 per cent collagenase overnight at 37°C to separate the epidermis.…”
