Fish Scale Collagen Peptides Protect against CoCl2/TNF‐α‐Induced Cytotoxicity and Inflammation via Inhibition of ROS, MAPK, and NF‐κB Pathways in HaCaT Cells

Abstract: Skin diseases associated with inflammation or oxidative stress represent the most common problem in dermatology. The present study demonstrates that fish scale collagen peptides (FSCP) protect against CoCl2-induced cytotoxicity and TNF-α-induced inflammatory responses in human HaCaT keratinocyte cells. Our study is the first to report that FSCP increase cell viability and ameliorate oxidative injury in HaCaT cells through mechanisms mediated by the downregulation of key proinflammatory cytokines, namely, TNF-α… Show more

“…Pemanfaatan ekstrak kolagen diantaranya untuk industri kosmetik, farmasi (penyembuh luka), dan makanan karena kolagen memiliki sifat daya tarik (tensile strength) yang tinggi, antigenisitas rendah, dan biokompatibilitas yang baik. Kolagen juga dapat menginduksi koagulasi trombosit, mempengaruhi diferensiasi sel, dan berkontribusi dalam penyembuhan luka (Subhan et al 2017;Dang et al 2017).…”

“…Much lower cytotoxicity activity in HaCaT cells was recorded as 19.9±1.9% at 1 mg/mL concentration by fish scale collagen peptide (FSCP) (Subhan et al, 2017). It is clear from the above studies that reduction of cell growth by collagen or collagen peptides are affected by the type of the cell, type of the fish and even degree of hydrolysis (DH).…”

“…Inhibition of relative cell growth was most likely not due to cell apoptosis or necrosis; and the related mechanism seems very complex Subhan et al (2017). As the hydrolysis of collagen induces collapse of the triple helix conformation, the obtained collagen peptides with different conformation influence the adhesion of molecules on the surfaces of different cancer cells.…”

“…Epidermal defense against the intrusion of harmful substances from the environment, including chemicals and radiation, acts as an important barrier to prevent skin injury (1,2). Keratinocytes are the principal cells of the epidermis.…”

“…Keratinocytes are the principal cells of the epidermis. In addition to their barrier-serving role, these cells are involved in immune responses within the skin; however, unperturbed keratinocytes exhibit deficient or absent production of inflammatory mediators (2). Under environmental or chemical stimuli (for example, UV irradiation or ambient air pollution), activated keratinocytes express numerous inflammation-associated cytokines including TNF-α, interleukin (IL)-1β, IL-8, and inducible nitric oxide synthase (iNOS), which may result in the abnormal expression and dysregulated action of inflammatory mediators (3,4).…”
