Fish oil administration mediates apoptosis of Walker 256 tumor cells by modulation of p53, Bcl-2, caspase-7 and caspase-3 protein expression

Abstract: BackgroundSeveral studies have been shown pro-apoptotic effects of fish oil (FO), rich in n-3 polyunsaturated fatty acids (n-3 PUFA) on cancer cells. Nevertheless, few in vivo experiments have provided data of its ability on apoptosis protein expression in tumor tissue. Thus, in this study we investigate the effect of FO supplementation on apoptosis protein expression in Walker 256 tumor bearing rats. Male Wistar rats were randomly assigned to three groups: fed with regular chow (W); fed regular chow supplemen… Show more

“…Bel-7402 cells treated with Plac1 siRNA showed decreased expression of Bcl-2, which was inversely related to p53 expression, implying that downregulation of Bcl-2 expression in Bel-7402 cells might be suppressed by p53. This hypothesis is supported by previous studies showing that Bcl-2 is negatively regulated by the tumor suppressor p53 (23,24). Surprisingly, instead of upregulation, Bax expression was inhibited in Bel-7402 cells transfected with Plac1 siRNA.…”

“…This translocation results in the release of cytochrome‐c into the cytosol, which forms a complex with factor activator of apoptosis 1 (Apaf‐1), activating caspases that subsequently trigger apoptosis . The caspase‐3 signalling mediating apoptosis in tumour cells has been described for several compounds, such as doxorubicin, eudesmol isomers , polyunsaturated fatty acids and polyacetylenes . For the first time, our data confirm the activation of caspase cascade by Syd‐1 in tumour cells.…”

“…It will be interesting and important to conduct a lifetime tracking on the growth rate, health status, and life span of Fat-1 transgenic sheep, as well as other Fat-1 transgenic livestock, to provide important references for humans under the condition of high ALA or n-3 LC-PUFA intake. Several reports indicated that n-3 PUFAs had an anticancer effect by activating the p53 pathway [52][53][54][55]. However, in our case, the p53 signaling pathway was also down-regulated in transgenic-expressing cells.…”
